去整合素Adinbitor对人胃癌细胞系MGC803细胞侵袭迁移影响的研究

【摘要】 目的:观察去整合素Adinbitor对人胃癌细胞系MGC803细胞侵袭和迁移能力的影响。方法:MGC803细胞经Adinbitor处理后,用结晶紫染色法检测其对纤维粘连蛋白(fibronectin,FN)和人工基质膜Matriger黏附能力的变化;聚碳酸酯膜浸润实验观察其侵袭和迁移能力的变化。结果:3.5μmol/L浓度的Adinbitor对MGC803细胞黏附FN的抑制率为44%;对黏附人工基底膜基质Matrigel也有明显的抑制作用,且呈剂量依赖性,F=144.31,P=0.000。迁移实验结果显示,1.1、3.3和5.5μmol/LAdinbitor组迁移细胞数分别为(78.00±2.36)、(57.00±1.41)、(33.50±1.52)个,与对照组(90.00±3.16)比较差异有统计学意义,F=743.59,P=0.000。侵袭实验结果显示,1.1、3.3和5.5μmol/LAdinbitor组浸袭细胞数为(72.00±2.61)、(60.17±3.19)、(38.00±2.61)个,与对照组(95.33±1.86)比较差异有统计学意义,F=503.47,P=0.000。结论:去整合素Adinbitor具有抑制MGC803细胞侵袭迁移的作用。

【Abstract】 OBJECTIVE:To study the influences of disintegrin Adinbitor on invasive and migrative abilities of human gastric carcinoma MGC803 cells. METHODS:MGC803 cells were treated with Adinbitor,then the adhesion abilities of MGC803 to fibronectin(FN) and Matrigel were detected respectively by crystal violet staining,and the ability of migration and invasion of MGC803 was assessed by the incorporation of polycarbonate filters in modified transwell chambers.RESULTS:Adinbitor in 3.5 μmol/L inhibited the adhesion of MGC803 cells to FN by 44%,and to Matrigel in dose-dependent manner (F=144.31,P=0.000). The numbers of migration cells and invasion cells decreased significantly by the treatment of Adinbitor in 1.1,3.3 and 5.5 μmol/L respectively (control of migration cells:90.00±3.16; treated by Adinbitor:78.00±2.36,57.00±1.41,33.50±1.52; control of invasion cells:95.33±1.86; treated by Adinbitor:72.00±2.61,60.17±3.19,38.00±2.61)(F=743.59,P=0.000; F=503.47,P=0.000).CONCLUSION:Disintegrin Adinbitor may inhibit the migration and invasion of MGC803 cells.
