双氢青蒿素对小鼠Lewis肺癌移植瘤生长的抑制作用
Effect of dihydroarteannuin on metastatic tumor growth in Lewis lung cancer bearing C57BL/6J mice
【摘要】 目的:探讨双氢青蒿素(DHA)对小鼠Lewis肺癌移植瘤的抑瘤效应及其作用机制。方法:C57BL/6J小鼠皮下接种3LL细胞(2×106)50只,随机分为5组,分别为生理盐水组、阳性对照顺铂(DDP)组和DHA高、中、低剂量组,检测各组小鼠的体质量变化和抑瘤率,应用流式细胞术进行瘤细胞的DNA倍体分析。结果:生理盐水组小鼠体质量比DHA高、中剂量增加明显,DHA低剂量抑瘤率为7.01%,较生理盐水组差异无统计学意义(t=0.872,P=0.395),中高剂量组抑瘤率分别为53.50%及59.24%,与生理盐水组比较差异有统计学意义(t=8.541,P=0.000;t=13.566,P=0.000),但抑瘤率均低于DDP组的92.36%,差异有统计学意义(t=-6.871,P=0.000;t=-9.667,P=0.000)。流式细胞术检测DHA能诱导肿瘤细胞凋亡,并能影响肿瘤的细胞周期,G0/G1期及G2/M期细胞数大量减少,细胞被阻滞在S1期,与生理盐水组比较,差异均有统计学意义,P值均为0.000。结论:在一定剂量范围内,口服DHA对Lewis小鼠肺癌有明显的抑制作用,可促进肿瘤细胞凋亡。
【Abstract】 OBJECTIVE:To explore the anti-tumor effect and the mechanism of dihydroarteannuin(DHA)in lung cancer bearing C57BL/6J mice.METHODS:Fifty C57BL/6J mice subcutaneous planted with 3LL lung cancer cells(2×106)were randomly divided into 5 groups:a blank control group with same volume of normal saline;positive control DDP group;low,middle and high dose groups.The body weight changes and inhibitory rate of the mice in each group were observed.In addition,the cell cycle and apoptosis index were analysed by flow cytometry.RESULTS:The body weights were decreased in the middle and high dose groups vs the NS group.Compare with the NS group,the inhibitory rate of the low dose group was 7.01%,there was non-significantly difference(t=0.872,P=0.395),and at the same time,in the middle and high dose groups the inhibitory rates were 53.50% and 59.24% respectively which were lower than that of DDP group(92.3%;t=-0.871,P=0.000;t=-9.667,P=0.000),and there were significant differences(t=8.541,P=0.000;t=13.566,P=0.000).The FCM assay indicated that DHA induced the apoptosis of lung cancer cells.At the same time,the number of G0/G1 and G2/M phase cells were significantly decreased,most of the cells were significantly arrested in S phase compared with the NS group,P=0.000.CONCLUSION:DHA is provided with the anti-tumor effect to lung cancer bearing C57BL/6J mice at certain range of dosage.Its mechanism might relate to inducing cancer cell apoptosis.
