参芪扶正注射液对荷瘤动物生存期影响的免疫机制研究

【摘要】 目的:探讨参芪扶正注射液对荷瘤动物细胞因子Th1/Th2平衡的调节作用,明确该药对生存期的影响及其可能机制。方法:以荷瘤Wistar大鼠为模型,分别给予参芪扶正注射液及顺铂,并以模型组及正常大鼠为对照。用流式细胞仪检测外周血Th1、Th2及其比值,同时用ELISA法检测外周血中IFN-γ和IL-4的含量。结果:化疗组和参芪扶正注射液组Th1/Th2值均高于模型组,P<0.01;但化疗组仍低于空白对照组,P<0.05。参芪扶正注射液组外周血IL-4较模型组和化疗组明显降低,P<0.01;而IFN-γ则明显提高,P<0.01。参芪扶正注射液组生存期较模型组和化疗组明显延长,P<0.01。结论:参芪扶正注射液可以增加Th1类细胞因子,减少Th2类细胞因子,使Th2向Th1漂移,这可能是该药延长荷瘤大鼠生存期的机制之一。

【Abstract】 OBJECTIVE: To explore the regulation on cytokines Th1/Th2 of Shenqi Fuzheng injection,and observe its effect on survival time and its possible mechanism.METHODS: Tumor bearing Wistar rats were given Shenqi Fuzheng injection and cisplatin respectively,and the model group and normal rats were taken as the control groups.Th1,Th2 and Th1/Th2 were tested by flow cytometry and the contents of IFN-γ and IL-4 in peripheral blood were tested by ELISA.RESULTS: Th1/ Th2 of the Shenqi Fuzheng injection group and cisplatin group increased greatly,P<0.01,but that in the cisplatin group still decreased significantly compared with the normal group,P<0.05.IL-4 of the Shenqi Fuzheng injection group decreased significantly compared with the model group and cisplatin group(P<0.01),but IFN-γ increased greatly,P<0.01.The survival time of the Shenqi Fuzheng injection group prolonged significantly compared with the model group and cisplatin group,P<0.01.CONCLUSIONS: Shenqi Fuzheng injection can increase Th1 cytokines and decrease Th2 cytodines,make Th2 shift to Th1.This might be one of the mechanisms of Shenqi Fuzheng injection prolonging the survival time of the tumor bearing rats.
