【摘要】 目的:探讨胃癌患者外周血单个核细胞(PBMC)中Th1和Th2类细胞因子mRNA表达变化及其临床意义。方法:采用RT-PCR技术,检测102例胃癌组织化疗前后、60例胃良性疾病组织和50例正常对照组织PBMC中Th1和Th2类细胞因子mRNA表达。结果:正常对照及胃良性疾病组PBMC中Th1和Th2类细胞因子基因阳性率差异无统计学意义,P>0.05。胃癌组PBMC中IL-4、IL-6和IL-10 mRNA阳性率显著高于正常对照和胃良性疾病组(P<0.05),且与胃癌的分化程度及临床分期相关,P<0.05;IFNγ-仅3例阳性表达(3/102),IL-2有7例阳性表达,显著低于正常对照和胃良性疾病组;胃癌组化疗有效者PBMC中Th1类细胞因子阳性率增加,Th2类细胞因子的阳性率则减低,P<0.05;化疗无效者PBMC中Th1和Th2类细胞因子mRNA阳性率较化疗前差异无统计学意义,P>0.05。结论:检测Th1和Th2类细胞因子mRNA的表达对评价胃癌患者的免疫状况和疾病进展及指导临床对患者进行免疫调节治疗有一定的应用价值。更多还原

【Abstract】 OBJECTIVE:To investigate the mRNA expressions of Th1 and Th2 cytokines in peripheral blood mononuclear cells(PBMC) from patients with gastric carcinoma.METHODS:The mRNA expressions of Th1/Th2 cytokines geng were detected by RT-PCR in 50 normal controls,60 cases of gastric benign diseases and 102 patients with gastric carcinoma before and after chemotherapy.RESULTS:The mRNA expressions of Th1 and Th2 cytokines had no significant difference in PBMCs from those between benign diseases and the normal controls,P>0.05.The mRNA expressions of IL-4,IL-6 and IL-10 in PBMCs from patients with gastric carcinoma were significantly higher than those from benign diseases and the normal controls(P<0.05),and correlated with the patients’ clinical stages and differential degrees,P<0.05.There were only 3 patients with a positive expression of IFN-γ(3/102) and 7 patients with a positive IL-2 mRNA expression in all the patients with gastric carcinoma,which were lower than those of benign diseases and the normal controls.The expression of Th1 was increased significantly,while that of Th2 was decreased in the group sentitive to chemotherapy of gastric carcinoma patients,P<0.05.There was no difference in the mRNA expressions of Th1 and Th2 in the group non-sensitive to chemotherapy,P>0.05.CONCLUSION:The detection of the gene expressions of Th1 and Th2 cytokines in PBMC from patients with gastric carcinoma is a sentitive and accurate means of evaluating the function of patient’s conditions and guiding the therapy.更多还原
