唐古特大黄细胞悬浮培养中的生理生化特性

【摘要】 目的初步研究液体培养条件下唐古特大黄(Rheum tanguticum Maxim.ex Balf.)细胞的生理生化特性.方法以唐古特大黄子叶为材料,诱导和筛选愈伤组织,进行细胞悬浮培养.测定了细胞悬浮培养过程中的细胞生长、pH值和总蒽醌累积的动态变化,并研究了水杨酸(SA)对细胞生长和总蒽醌累积的影响.结果培养细胞生长周期36d;细胞的增殖曲线呈"S"形;其培养液pH值先下降后回升;细胞中总蒽醌累积规律也呈"S"形曲线,在培养的第30天,细胞中总蒽醌可达到最大值.SA对细胞的生长和总蒽醌累积有影响,0.1mg/L和1.0mg/L SA促进细胞的生长和总蒽醌的积累.结论细胞悬浮培养过程中的总蒽醌累积和细胞生长呈正相关性.

【Abstract】 Objective To primarily study the physiological and biochemical characters of Rheum tanguticum Maxim.ex Balf.suspension culture.Methods With the cotyledon of Rheum tanguticum Maxim.ex Balf.callus was induced and screened,then established the suspension culture,the dynamic changes of cell growth,pH and anthraquinones accumulation were monitored.The effects of salicylic acid(SA) on cell growth and anthraquinones accumulation were studied.Result The cell growth curve was like "S",but the growth circle was about 36 days;the pH of cultured mediums went down at first,then went up gradually;the anthraquinones accumulation curve in suspension cell of Rheum tanguticum Maxim.ex Balf.was also like"S";the anthraquinones content in suspension cell was highest when suspension cell were cultured in the 30th day.The SA had some effects on the cell growth and anthraquinones accumulation in suspension cell of Rheum tanguticum Maxim.ex Balf.,0.1mg/L of SA and 1mg/L of SA could promote the growth of Rheum tanguticum Maxim.ex Balf.cell and the anthraquinones accumulation.Conclusion There was a plus relativity between cell growth and anthraquinones accumulation.
