【摘要】 目的探讨慢性心力衰竭大鼠心肌内半胱氨酸天冬氨酸蛋白酶3(caspase-3)表达及意义。方法40只雄性Wistar大鼠随机分为四组,假手术组、心力衰竭1d组、7d组、14d组各10只。以缩窄大鼠腹主动脉,造成左心室压力负荷,制备慢性心力衰竭大鼠模型。观察和比较成模后1d、7d、14d及假手术组左心室肌重量指数,Masson染色后对比心肌胶原含量,免疫组化法测定心肌中caspase-3蛋白表达水平。结果腹主动脉缩窄术后各组大鼠均发生左室重塑和纤维化,左心室肌重量指数、胶原含量及caspase-3蛋白表达均明显高于假手术组;caspase-3于正常心肌内表达较少,而在心力衰竭后心肌内表达则明显增多,且随着心力衰竭程度的加重而表达增多。结论caspase-3可能参与了大鼠压力负荷性心肌重塑的发生和发展,其水平的高低与慢性心力衰竭的严重程度密切相关。更多还原

【Abstract】 Objective To investigate the dynamic expression and significance of caspase-3 in rats of heart failure induced by pressure overload.MethodsThe male Wistar rats were randomly divided into the sham-operation group,heart failure group 1 (for one day),heart failure group 2 (for 7 days) and heart failure group 3 (for 14 days,each n=10). The rat model of chronic heart failure was established by coarctation of abdominal aorta for creating pressure overload in the left ventricle. The left ventricular mass indexes were observed in all groups. The myocardial collagen contents were compared after Mason staining,and the expression of caspase-3 was detected by immunohistochemical method.ResultsThe left ventricle remodeling and fibrosis were observed in all groups. The left ventricular mass indexes,collagen contents and expression of caspase-3 were higher significantly in heart failure group 1,2 and 3 than those in the sham-operation group. The expression of caspase-3 was lower in normal cardiac muscle but higher after the onset of heart failure,and more higher along with the aggravation of heart failure.Conclusioncaspase-3 may be involved with the development of myocardial remodeling in rats of pressure overload,and its level is related closely to the severity of chronic heart failure.更多还原
