【摘要】 目的探讨蛋白激酶C(protein kinase C,PKC)-α在肾癌及其周围正常组织胞质和胞膜中的表达和意义以及与肾癌分期、分级的关系。方法采用Western blotting技术检测30例肾癌组织及其周围正常组织胞质、胞膜中PKC-α的表达。结果在肾癌组织或邻近正常组织的胞质和胞膜中PKC-α都有一定量的表达。与邻近正常组织相比,肾癌组织胞质中PKC-α表达降低,胞膜中PKC-α表达增高,膜/质比上升,差异均有统计学意义(P<0.01)。随着肾癌病理分期和组织学分级的提高,PKC-α在肿瘤组织胞膜的表达也相应增高,在肿瘤组织胞质的表达则相应降低,膜/质比增加,差异均有统计学意义(P<0.01)。结论PKC-α在肾癌发生、发展过程中可能起了重要的调控作用;PKC-α在肾癌组织中存在膜转位;PKC-α在肾癌组织胞膜中高表达,在肾癌组织胞质中低表达,并且与肾癌的病理分期和组织学分级密切相关;PKC-α是判断肾癌分化、预后的一种理想指标。更多还原

【Abstract】 Objective To investigate the expression and significance of PKC-α in membrane and plasma of renal cell carcinoma and adjacent normal tissues,and the relationship between PKC-α and stages and grades of renal cell carcinoma has been analyzed. Methods The expression of PKC-α in membrane and plasma of 30 cases renal cell carcinoma and normal adjacent tissues has been investigated by Western blotting technology. Results There is expression of PKC-α in membrane and plasma of renal cell carcinoma or normal adjacent tissues.Compared to that of adjacent normal tissues,the expression of PKC-α in plasma is decreased significantly,while its expression in membrane is increased significantly and the ratio of membrane/plasma increases significantly(P<0.01).The difference has statistical significance.In membrane of renal cell carcinoma tissue,the expression of PKC-α is increased with the elevation of pathological stages and cell grading of tumor(P<0.01),while in plasma of tumor,the expression of PKC-α is decreased with the elevation of pathological stages and cell grading of tumor(P<0.01),and the ratio of membrane/plasma increases significantly. Conclusions PKC-α maybe play an important regulative role in the occurrence and development process of renal cell carcinoma.Membrane transposition of PKC-α is present in renal cell carcinoma tissue.The expression of PKC-α increases in membrane of tumor and decreases in plasma,and concerns with the grade and stage of renal cell carcinoma closely.PKC-α is one of the ideal indexes to judge the differentiation and prognosis of renal cell carcinoma.更多还原
