整合素β1、VEGF及MMP-9在膀胱移行细胞癌中的表达及意义
The expression and significance of integrinβ1,VEGF,MMP-9 in bladder transitional cell cancer
【摘要】 目的检测整合素β1、血管内皮生长因子(VEGF)及基质金属蛋白酶-9(MMP-9)在人膀胱移行细胞癌的表达,探讨整合素β1与膀胱移行细胞癌血管生成及浸润转移的关系。方法采用免疫组织化学SABC法检测59例膀胱移行细胞癌组织及15例正常膀胱黏膜组织中整合素β1、VEGF、MMP-9的表达。结果59例膀胱移行细胞癌中整合素β1、VEGF、MMP-9的阳性表达率分别为45.76%、64.41%和55.93%,15例正常膀胱黏膜组织中三者的阳性表达率分别为13.33%、6.67%和6.67%,膀胱移行细胞癌与正常膀胱黏膜组织间3种蛋白阳性表达率差异均有统计学意义(P<0.05,P<0.01,P<0.01);整合素β1、VEGF、MMP-9的表达与膀胱移行细胞癌的分期、分级相关;整合素β1、VEGF的表达与膀胱移行细胞癌的癌灶直径、单发/多发相关;整合素β1与膀胱移行细胞癌患者的年龄相关;整合素β1与VEGF及MMP-9之间的阳性表达均呈正相关。结论整合素β1在膀胱移行细胞癌中表达升高,并与分级、分期等临床病理指标相关,表明其具有促进膀胱移行细胞癌生长的作用。整合素β1可能通过协同VEGF及MMP-9作用促进膀胱移行细胞癌的血管生成及侵袭转移。
【Abstract】 Objective To find out how integrinβ1 play a role in the angiogenesis,invasion and metastatic of bladder transitional cell cancer(BTCC),through detecting the expression of integrinβ1,VEGF,MMP-9 in BTCC.Methods Using the SABC kit to detect the expression of integrinβ1,VEGF,MMP-9 in 59 cases of BTCC paraffin tissues collected from the patients underwent surgical resection,and 15 cases of normal bladder tissues from the patients underwent the resection of benign prostatic hyperplasia as negative control.Results The positive expression percentage of integrinβ1,VEGF,MMP-9 in BTCC was 45.76%,64.41%,55.93% respectively,and the positive expression percentage in normal bladder tissues was 13.33%,6.67%,6.67% respectively.The difference between two groups was significant(P<0.05,P<0.01,P<0.01).The expression of integrinβ1,VEGF,MMP-9 had relation with stage and grade.The expression of integrinβ1 and VEGF had relation with simple/poly disease and diameter of tumor.The expression of integrinβ1 also had relation with age of patients.There was a positive correlation between the expression of integrinβ1 and VEGF or between integrinβ1and MMP-9.Conclusions The expression of integrinβ1 increases in BTCC,and has relation with the clinicopathlogical index such as stage or grade,integrinβ1 might have cooperation with VEGF and MMP-9 in the angiogenesis,invasion and metastatic of BTCC.
