主动脉夹层中硫酸软骨素含量和结构改变的初步研究

【摘要】 目的:比较正常主动脉壁和主动脉夹层瘤壁中硫酸软骨素的含量及结构,探讨硫酸软骨素在主动脉夹层发生和进展中的可能作用。方法:收集主动脉夹层瘤壁标本(实验组)10例和正常主动脉壁标本(正常对照组)7例,提取两组标本中的糖胺聚糖,采用DEAE Sephacel离子交换层析法分离纯化硫酸软骨素。以硫酸软骨素酶ABC将硫酸软骨素裂解为不饱和二糖,应用高效毛细管电泳法进行测定。结果:实验组中硫酸软骨素含量较正常对照组明显升高(P<0.01)。实验组中硫酸软骨素二糖包括Δdi-di(2,6)S、Δdi-di(2,4)S、Δdi-di(4,6)S、Δdi-mono6S、Δdi-mono4S和Δdi-nonS,其中除Δdi-di(2,4)含量较正常对照明显降低(P<0.05)外,其余5种二糖均明显升高(P<0.05)。结论:主动脉夹层瘤壁中硫酸软骨素的含量和分子结构均发生明显异常。这些异常可能在主动脉夹层的发病和进展过程中发挥重要作用。

【Abstract】 Objectives:To elucidate the changes on contents and molecular structures of chondroitin sulfate in aneurysmal wall of aortic dissection and disscuss its likely roles in the pathogenesis and progression of aortic dissection.Methods:After glycosaminoglycans in aortae being abstracted,chondroitin sulfate were separated and purified respectively through DEAE Sephacel by ion exchange chromatography and then identified by cellulose acetate electrophoresis.Chondroitin sulfate were hydrolyzed with chondroitinase ABC,and then its disaccharide compositions were analyzed by high performance capillary electrophoresis.Results:Compared with the control group, chondroitin sulfate increased significantly(P<0.01) in the experimental group.Disaccharide compositions of chondroitin sulfate in the experimental group were detected to contain△di-di(2,6)S,△di-di(2,4)S,△di-di(4,6)S,△di-mono6S,△di-mono4S and△di-nonS by high performance capillary electrophoresis,of which△di-di(2,4)S decreasing significantly(P<0.01) while the other five kind increasing significantly(P<0.01).Conclusion:Both contents and molecular structures of chondroitin sufate changed significantly in aneurysmal wall of aortic dissection,which may lead to weakness of the aortic wall and damage the functional integrity of the aorta.We consider that abnormal contents and molecular structures of chondroitin sufate may play a key role in the initialization and progression of aortic dissection.
