【摘要】 目的:研究胆囊黏膜ABCG5和ABCG8的基因表达与胆囊胆固醇息肉发病的关系。方法:收集63例因胆囊疾患而行腹腔镜胆囊切除术患者的胆石、胆汁及部分胆囊黏膜组织,其中胆囊胆固醇息肉32例,胆囊胆固醇结石18例,对照13例(胆囊腺瘤6例,非胆固醇胆囊结石7例),分别测定胆石胆固醇含量,胆汁胆固醇、胆汁酸、磷脂的浓度,实时定量PCR检测胆囊黏膜ABCG5和ABCG8的mRNA表达。结果:胆固醇息肉组胆汁胆固醇饱和指数较对照组明显增高(P<0.01),结石组胆囊黏膜ABCG5和ABCG8的表达量较息肉组和对照组显著增高,息肉组和对照组ABCG5和ABCG8的表达量差异无统计学意义(P>0.05)。结论:胆囊黏膜ABCG5/ABCG8mRNA的相对低水平表达,可能是导致胆囊胆固醇息肉发病的重要因素之一。更多还原

【Abstract】 Objective:To study the relationship between the gene expression of ABCG5 and ABCG8 in gallbladder mucosa and the pathogenesis of gallbladder cholesterol polyps. Methods:Gallstone,bile and gallbladder mucosa were collected from 63 patients who underwent laparoscopic cholecystectomy,including 32 patients with gallbladder cholesterol polyps,18 patients with cholesterol gallstones and 13 patients in control(6 patients with gallbladder adenoma and 7 patients with non-cholesterol gallstone). The contents of cholesterol in gallstone and cholesterol,bile acid and phospholipid in bile were measured.The mRNA expression of ABCG5 and ABCG8 in gallbladder mucosa were quantified with real-time PCR. Results:Cholesterol saturation index was significantly higher in the patients with cholesterol polyps compared with that in control group. The mRNA expression of ABCG5 and ABCG8 were significantly higher in gallbladder mucosa from the patients with cholesterol gallstone compared with those that in either patients with gallbladder polyps or the control group. However,there was no difference of the mRNA expression of ABCG5 and ABCG8 between the patients with gallbladder polyps and control group. Conclusion:Relatively lower level mRNA expression of ABCG5 and ABCG8 in gallbladder mucosa may be one of the important factors of the pathogenesis of gallbladder cholesterol polyps.更多还原
