高效液相色谱法测定人血浆中芬太尼的浓度测定分析
Determination of concentration of fentanyl in human plasma by HPLC
【摘要】 目的建立高效液相色谱(HPLC)法测定芬太尼血药浓度的方法。方法本实验以乙腈∶磷酸盐(0.015ol/L NaH2PO4,pH=5)=40∶60为流动相,流速3ml/min,紫外吸收波长为200nm。结果高效液相色谱法对血浆中药物的最低检测浓度为1ng/ml,线形范围6-300ng/ml。对芬太尼方法回收率均大于95%,日间日内相对标准偏差均小于10.0%。结论高效液相色谱法简便、准确,能满足临床对芬太尼血药浓度的测定。
【Abstract】 Objective To establish a HPLC method for determination of concentration of fentanyl in human plasma.Methods The mobile phase was composed of acetonitril-water(0.0015 mol/LNaH2PO4,pH=5.0) =40∶60,the UV detector wavelength was at =200 nm.Results The detection limit was 1ng/ml,and the calibration curve was linear in the range from 6-300ng/m1.The recovery of fentanyl from human plasma was more than 95%.The daily and intraday and RSD were all less than 10.0%.Conclusion The HPLC method is rapid and accurate for determination of fentanyl in human plasma.
