蒙药娜仁满都拉-11对种植性胃肿瘤鼠IL-4和IFN-γ调节作用研究

【摘要】 目的:研究娜仁满都拉-11的抗肿瘤作用及其对IL-4、IFN-γ的免疫调节作用,为进一步开发该药提供实验依据。方法:建立Walker-256种植性胃肿瘤大鼠模型,用娜仁满都拉-11灌胃10 d后,取瘤组织称重计算抑瘤率;从心脏血中分离单个核细胞进行细胞培养,用酶联免疫吸附法(ELISA),RT-PCR法检测IL-4、IFN-γ的表达水平。结果:瘤重抑制率为44%;该药治疗组动物IL-4水平显著低于阴性对照组,而IFN-γ水平显著高于阴性对照组。结论:娜仁满都拉-11能抑制Walker-256种植性胃肿瘤生长,并具有一定的抗肿瘤免疫调节作用,增强IFN-γ的表达,降低IL-4的表达。

【Abstract】 Objective:Study the anti-tumur effect of Na Ren Man Du La-11 and influence of IL-4,IFNγ on immune adjustment,provide evidence for the further study of developing Na Ren Man Du La-11.Method: Walker-256 cell lines were used to duplicate implanted tumor model in rat,different dose of Na Ren Man Du La-11 were given to the rat in treatment groups,10 days later we get out tumor tissue and test their weights,then separate mononuclear cell from heart blood and cultivated them.ELISA and RT-PCR were used to test quantity the expression of IL-4 and IFN-γ.Results: Implanted gastric tumor can be inhibited by Na Ren Man Du La-11 in some degree,Weight-inhibit rate is 44%.IL-4 is lower in treatment group than that in negative control group,while IFN-γ is higher than that in control group.Conclusion: Na Ren Man Du La-11 has anti-tumor function in treating implanted tumor of Walker-256 cells on the animal model, it also has function in immune adjustment by enhancing the expression of IFN-γ and lower expression of IL-4.
