硫酸化黄芪多糖对CEF生长及抵抗IBDV感染的影响
Effect of sulfated astragalus polysaccharides on growth of chicken embryo fibroblast and cellular infectivity of infectious bursal disease virus
【摘要】 采取一步醇沉和分步醇沉法提取4种黄芪多糖APSt、APS40、APS50和APS60,用氯磺酸-吡啶法进行硫酸化修饰,得到4种硫酸化黄芪多糖(sAPS)sAPSt、sAPS40、sAPS50和sAPS60,用MTT法比较了4种硫酸化黄芪多糖对鸡胚成纤维细胞(CEF)的生长及抵抗鸡传染性法氏囊病毒(IBDV)感染的影响,以未修饰的APSt为对照。结果显示,APSt显著促进CEF生长,细胞毒性较低,但抵抗鸡传染性法氏囊病毒感染细胞的作用较弱;sAPS抵抗鸡传染性法氏囊病毒感染的作用显著增强,显效浓度降低,但安全浓度也降低。表明通过硫酸化修饰可以提高黄芪多糖的抗病毒活性,但高浓度sAPS的细胞毒性增强。
【Abstract】 Four kinds of astragalus polysaccharides(APS),APSt,APS40,APS50 and APS60,were extracted by one-step and stepwise ethanol precipitation method and modified by chlrulfonic-pyridine method to obtain four sulfated APS(sAPS),sAPSt,sAPS40,sAPS50,sAPS60,respectively.The effects of four sAPS on growth of chicken embryo fibroblast(CEF)and cellular infectivity of infections bursal disease virus(IBDV)were compared by MTT method taking non-modified APSt as control.The results showed that non-modified APSt promoted CEF growth significantly and presented lower cytotoxicity,but its activity of resisting IBDV to infect CEF was weaker than sAPS.However,modified sAPS inhibited IBDV to infect CEF significantly and effective concentration were lowered,but their cytotoxicities became stronger than APSt at high concentration.These results indicated that sulfated modification could enhance the antiviral activity of APS and cytotoxicity of sAPS at high concentration.
