硫酸铵盐析法分离罗非鱼肌红蛋白的研究

【摘要】 通过测定盐析液的色泽、肌红蛋白含量、纯度、回收率和吸收光谱等的变化情况,分析了硫酸铵分级盐析过程对罗非鱼肌红蛋白分离的影响。研究表明,硫酸铵饱和度为60%~100%可导致肌红蛋白沉淀析出,其中饱和度为50%时,肌红蛋白纯度为37.52%,回收率仅为39.89%;饱和度大于60%时,肌红蛋白纯度虽有所降低,但回收率逐渐提高,当饱和度为100%时,回收率最高为87.56%。在盐析过程中,部分氧合肌红蛋白氧化成为高铁肌红蛋白。

【Abstract】 Effect of ammonium sulfate on salting-out of myoglobin was studied by analysing color,Mb content,Mb purity,recovery rate and absorption spectra.The results showed that the myoglobin could be precipitated after treating with ammonium sulfate at saturation 60%~100%,and the purity and recovery rate were 37.52% and 39.89% respectively after treated with 50% ammonium sulfate.Mb purity was slightly decreased but the recovery rate was increased gradually when the saturation was greater than 60%.And recovery rate was up to 87.56% when the saturation reached to 100%.Oxymyoglobin was partly oxidated into the met-myoglobin in the process of the ammonium sulfate salting-out.
