套管构建肾血管性高血压大鼠模型
Establishment of Renovascular Hypertensive Rat Model by Cannula
【摘要】 为了探讨采用套管构建2 K1C肾血管性高血压大鼠模型的可行性,本实验选取SD大鼠60只,分为对照组、假手术组、2 mm手术组及3 mm手术组,两个手术组采用内径为0.3 mm,长度分别为2 mm和3 mm两种自制不锈钢金属套管,对大鼠左肾动脉进行狭窄,构建2 K1C肾血管性高血压大鼠模型并使用尾容积法测量大鼠血压。结果如下:术前各组血压为14.93~15.04 kPa。术后对照组及假手术组血压与术前相比无显著性差异(P>0.05)。2 mm手术组和3 mm手术组,术后一周血压分别为18.20±1.21 kPa和17.96±1.68 kPa,各自与术前相比有极显著性差异(P<0.01),以上两个手术组血压持续上升至术后4周,之后趋于稳定,术后6周血压分别为26.39±1.79kPa和26.21±2.4 6kPa。术后6周对两个手术组解剖观察,发现大鼠左侧肾脏未发生明显萎缩,左肾动脉完好,套管未脱落。上述结果表明:采用套管对大鼠左肾动脉进行狭窄,可成功构建2 K1C高血压大鼠模型,一周后大鼠即可发生高血压,且血压急剧上升至术后4周趋于稳定。短期而言,套管的长度对高血压的形成及维持无显著性影响。
【Abstract】 In order to investigate the feasibility of establishing two-kidney one-clip renovascular hypertensive rat model by cannula,60 SD rats were divided into four groups:control group,sham group,2 mm operated group and 3 mm operated group.The left renal arteries of the two operated groups were narrowed respectively by the 2 mm or 3 mm long self-made stainless metal cannula of 0.3 mm inner diameter for establishing the 2 K1C hypertensive rat models.BP was measured by tail volume method.The results showed that the BP of all groups was 14.93~15.04 kPa before operation.After operation,there was no significant difference(P>0.05) in BP of the control group and the sham group compared with themselves before operation.The BP of the 2mm operated group and the 3mm operated group was 18.20±1.21 kPa and 17.96±1.68 kPa in one week after operation and there was significant difference(P<0.01)compared with themselves before operation.The BP of the two operated groups rose continuously till the four weeks after the operation and then tended to be stable,and six weeks after operation BP was 26.39±1.79 kPa and 26.21±2.46 kPa.The rats in two operated groups were dissected at the end of six weeks after operation and it was observed that the left kidneys did not develop obvious atrophy,left renal arteries were intact and the cannulas were still around the left renal arteries.That of results suggested that it was feasible to establish the 2 K1C hypertensive rat model by narrowing the left renal artery with cannula.The rat model formed hypertension in one week after operation and the BP increased sharply until the four weeks after operation and then became steady and stayed high level.In short-duration,there was no significant effect in the different length cannulas on the formation and maintenance of hypertension.
