B7在血液病细胞系中的表达及对T淋巴细胞的免疫反应

【摘要】 目的探讨B7共刺激分子在人类恶性血液病细胞系中的表达以及激活T淋巴细胞的免疫反应。方法用流式细胞术检测HL-60、NB4、Jurkat、U937、Raji、Daudi、K562等细胞HLA抗原和B7分子表达,同时检测经Ara-C刺激前后B7分子的表达,用RT-PCR方法检测经Ara-C刺激前后激活T淋巴细胞分泌细胞因子IFN-γ mRNA。结果HL-60、NB4、Jurkat、U937、Raji、Daudi、K562等恶性血液病细胞株均表达或高表达HLA抗原和B7-2分子,低表达或不表达B7-1分子,经Ara-C刺激后细胞株B7分子的表达明显增强,并能有效地激活T淋巴细胞表达高水平的IFN-γ mRNA。结论大多数人类恶性血液病细胞株均不表达或低表达B7分子,但是B7-1可能在肿瘤免疫中起重要作用。

【Abstract】 Objective To explore the expression of B7 co-stimulator in human malignant hematological cell lines and its effects on activation of human peripheral blood T lymphocyte. Methods The expressions of B7 and HLA molecules on fresh hematological cell lines and on the Ara-C exposed hematological cell were detected by FAC scan cytometer. IFN-γ mRNA in Ara-C treated hematological cells was detected by reverse transcription PCR (RT-PCR). Results Human malignant hematological cell lines HL-60,NB4,Jurkat,U937,Raji,Daudi,and K562 were all expressed high levels of HLA molecules and B7-2 gene,but low or undetectable level of B7-1 gene. Ara-C treated hematological cells could stimulate the proliferation of peripheral blood mononuclear cells and promote the IFN-γ production. Conclusion Multiple human malignant hematological cell lines express low or undetectable level of B7 gene,suggesting that B7-1 plays a critical role in tumor immunity.
