龙须菜对小鼠脾脏淋巴细胞内TGF-β表达的影响

【摘要】 目的研究大型海藻龙须菜提取物对小鼠脾脏淋巴细胞内TGF-β表达的影响,并初探其免疫调控作用机制。方法以大型海藻龙须菜为实验材料,采用流式细胞术对Balb/c小鼠脾脏淋巴细胞进行表型分析,检测了不同浓度龙须菜提取物共培养条件下CD45+细胞内转化生长因子(TGF-β)表达水平的变化,并对产生TGF-β的细胞进行身份鉴定。结果龙须菜提取物不同反应浓度处理条件下Balb/c小鼠脾脏淋巴细胞中CD45+CD83+/CD45+及CD45+CD80+/CD45+细胞百分率与对照组比较无显著性差异(P>0.05),但CD45+TGF-β/CD45+细胞百分率与龙须菜提取物反应液浓度则呈明显的线性剂量依赖关系(r2=0.8145,P<0.05),最适反应相对百分浓度为2%。TGF-β主要来源于CD3+T细胞(约占35.4%)和CD49b+NK细胞(约占46.6%)。结论提示龙须菜提取物具有通过TGF-β进行免疫抑制或生物调节的功能。

【Abstract】 Objective To explore the effects of Gracilaria lemaneiformis extraction on the expression of transforming growth factor β(TGF-β)in the splenic lymphocytes of Balb/c mice and to investigate the immunomodulatory mechanisms of the seaweed.Methods Lymphocyte phenotyping was performed with flow cytometry.The expression of TGF-β in CD45+ cells was detected under Gracilaria lemaneiformis extraction of different concentrations and those TGF-β producing cells were identified.Results No statistical difference in CD45+CD83+/CD45+ and CD45+CD80+/CD45+ cell percentage was observed between the Balb/c mice treated with Gracilaria lemaneiformis extraction of different concentrations and the control group(P>0.05).The CD45+TGF-β/CD45+ rate was obviously related with the seaweed extraction concentration in a dose-dependent manner(r2=0.814 5,P<0.05),and the best relative concentration of Gracilaria lemaneiformis extraction for reaction was 2%.TGF-β was mainly derived from CD3+ T cells and CD49b+ NK cells,accounting for 35.4% and 46.6%,respectively.Conclusion The research indicts the effective components of the seaweed have different immunomodulatory effects.
