多酚氧化酶活性基因等位变异在陕西小麦品种中的分布

【摘要】 为了解陕西小麦品种控制多酚氧化酶(Polyphenol oxidase,PPO)活性的主效基因组成,利用PPO18和PPO29标记对138份推广品种的PPO-2A和PPO-2D位点的等位变异进行了分子检测。结果表明,在PPO-2A位点,含Ppo-A1a(高PPO活性)和Ppo-A1b(低PPO活性)等位变异类型的品种比例分别为47.1%和52.9%;在PPO-2D位点,含Ppo-D1a(中等低PPO活性)和Ppo-D1b(中等高PPO活性)等位变异类型的品种频率分别为30.4%和69.6%;含Ppo-A1a/Ppo-D1a(较高PPO活性)、Ppo-A1a/Ppo-D1b(最高PPO活性)、Ppo-A1b/Ppo-D1a(最低PPO活性)、Ppo-A1b/Ppo-D1b(较低PPO活性)等位变异组合类型品种的频率依次为10.1%、37.0%、20.3%和32.6%;不同地区不同等位变异类型及其组合的分布比例也不同。总体来看,陕西低PPO活性的小麦品种比例偏低,选育低PPO活性品种应成为当前陕西小麦品质改良的主要目标之一。

【Abstract】 Colour is an important trait exerting significant influences on the appearance of wheat-based products.In order to investigate content of polyphenol oxidase(PPO) activity and presence of allelic variations at PPO-2A and PPO-2D loci in Shaanxi wheat cultivars,a total of 138 cultivars were investigated using functional markers of PPO 18 and PPO 29.The results indicated that the frequencies of cultivars with alleles Ppo-A1a(high PPO activity),Ppo-A1b(low PPO activity) at PPO-2A locus were 47.1% and 52.9%,respectively,and Ppo-D1a(low PPO activity) and Ppo-D1b(high PPO activity) at PPO-2D locus were 30.4% and 69.6%,respectively.The frequencies of allelic combinations of PPO genes for Ppo-A1a/Ppo-D1a(higher PPO activity),Ppo-A1a/Ppo-D1b(the highest PPO activity),Ppo-A1b/Ppo-D1a(the lowest PPO activity) and Ppo-A1b/Ppo-D1b(lower PPO activity) at PPO-2A and PPO-2D loci were 10.1%,37.0%,20.3% and 32.6%,respectively.The frequencies of those genotypes were also different among wheat cultivars from different regions of Shaanxi.Generally,frequency of cultivars with the lowest PPO activity in Shaanxi province was low.Therefore,an object of wheat quality improvement in Shaanxi province in the future would be to breed cultivars with lower PPO activity.
