小麦WVp-1基因表达载体的Gateway技术构建及其遗传转化
Construction and Transformation of the Expression Vector of Wheat WVp-1 Gene Using Gateway Technology
【摘要】 为了利用中国特有的抗穗发芽小麦遗传资源,并深入分析Viviparous-1(Vp-1)基因在小麦穗发芽抗性中的作用,从抗穗发芽小麦品种西农979的干种子中分离了Vp-1基因的同源基因,命名为WVp-1。利用Gate-way技术,以植物表达载体pLeela为基础,成功构建了含有双35S启动子的高效表达载体,并转化拟南芥突变体abi3-4,获得了8株阳性植株。
【Abstract】 Pre-harvest sprouting(PHS) of grain while it is still in the ear,is due to an early disruption of seed dormancy.PHS reduces the quality of wheat and the value of the grain.Viviparous1(Vp-1) homologues play an important role in seed maturation,dormancy and desiccation.In this study,to make use of genetic resource of pre-harvest sprouting tolerance in Chinese wheats,PCR-based approach was employed to isolate a Vp-1 homologous cDNA clone from Xinong 979,and designated as WVp-1.Using Gateway cloning technology,the plant expression vector of WVp-1 gene driven by the double 35S promoter was successfully constructed and induced into Arabidopsis mutant abi3-4.8 plants with resistance to Bialaphos were obtain.This laid a basis for the function analysis of WVp-1 gene.
