端粒酶活性对T2DM大鼠血管平滑肌细胞增殖的影响

【摘要】 目的观察2型糖尿病(T2DM)大鼠血管平滑肌细胞端粒酶活性及其对血管平滑肌细胞增殖的影响。方法应用高脂、高糖饮食以及腹腔注射小剂量链脲佐菌素的方法建立2型糖尿病大鼠模型,喂养1个月后检测大鼠血糖、血脂和血清胰岛素变化,收集胸主动脉平滑肌细胞,采用TRAP-ELISA法测量血管平滑肌细胞端粒酶活性,MTT法测定平滑肌细胞增殖程度。结果与对照组相比,2型糖尿病大鼠血糖、血脂和血清胰岛素均明显增高,血管平滑肌细胞端粒酶活性和平滑肌细胞增殖也增高(P<0.05),相关分析显示两者之间存在正相关关系(r=0.959,P<0.01)。结论 2型糖尿病大鼠血管平滑肌细胞端粒酶的活性明显增高,并与血管平滑肌细胞的增殖程度呈正相关,因此,端粒酶抑制剂有可能成为治疗2型糖尿病血管并发症的新途径。

【Abstract】 Objective To study the variety of telomerase activity and its influence on vascular smooth muscle cells proliferation in type 2 diabetes mellitus(T2DM).Methods The model rats were given high-energy and high-fat diet,with small dose of streptozotocin(STZ,30mg/kg) through intraperitoneal injection.Ten normal rats was selected as the control group.After feeding a month,the blood glucose,blood lipids and serum insulin level of the rat model were detected. At the same time,thoracic aortic smooth muscle cells were collected.Telomerase activity in vascular smooth muscle cells was detected by TRAP-ELISA.The degree of smooth muscle cells proliferation was measured by MTT.Results Compared with the control group,the rats in model group had high blood glucose,blood lilpid and serum insulin.The expression of telomerase activity and the degree of vascular smooth cells proliferation in model group were higher than that of the control group.Correlation analysis showed a positive correlation between them (P<0.05).Conclusion In the vascular smooth muscle cells of T2DM rats,the telomerase activity was increased.There was a positive correlation between the activity and proliferation of vasucular smooth muscle cells.This animal experiment offers a new way to further study the complications of T2DM.
