原癌基因c-met和半乳糖凝素-3在甲状腺肿瘤中的表达
Expression of proto-oncogene c-met and galectin-3 in thyroid neoplasm and its significance
【摘要】 目的探讨原癌基因c-met和半乳糖凝素-3(Gal-3)在甲状腺肿瘤中的表达特点及在鉴别诊断中的价值。方法采用PV-6000通用型免疫组织化学二步法检测正常甲状腺组织10例、结节性甲状腺肿15例、甲状腺腺瘤25例、甲状腺滤泡癌20例、甲状腺乳头状癌31例、甲状腺未分化癌2例中c-met和Gal-3的表达。结果c-met和Gal-3在滤泡癌、乳头状癌及未分化癌中的阳性表达率高于甲状腺腺瘤、结节性甲状腺肿。c-met表达的强阳性率与淋巴结转移有关,Gal-3表达的强阳性率与甲状腺癌组织分化程度、临床分期、淋巴结转移有关。癌旁正常甲状腺组织中c-met和Gal-3表达均为阴性。甲状腺癌c-met的阳性表达与Gal-3的阳性表达呈正相关关系。结论c-met和Gal-3过表达与甲状腺肿瘤的发生、发展密切相关,对甲状腺肿瘤的鉴别以及预后的评估有一定的价值。
【Abstract】 Objective To investigate the expression of proto-oncogene c-met and glactin-3(Gla- 3) oncogene protein in thyroid neoplasm and to assess their value as markers in the differential diagnoses.Methods Immunohistochemical PV-6000 technique was used to examine c-met and Gla-3 expression in 10 cases of normal thyroid tissue,15 cases of nodular goiter,25 cases of thyroid adenoma,20 cases of follicular carcinoma,31 cases of papillary carcinoma and 2 cases of undifferentiated carcinoma.Results The positive expression rates of c-met and Gla-3 in follicular carcinoma,papillary carcinoma and undifferentiated carcinoma were significantly higher than those of thyroid adenoma and nodular goiter.The strong positive rate of c-met expression was correlated with the metastasis of lymph nodes.The strong positive rates of Gla-3 expression were closely related with the tissue differentiation degree,clinical stage and lymph nodes metastasis in thyroid carcinoma.c-met and Gla-3 were negative in normal thyroid tissue.c-met positive expression in thyroid carcinoma was thus in positive correlation with the expression of Gla- 3.Conclusion The over-expression of c-met and Gla-3 are closely correlated with occurrence and development of thyroid neoplasm.The detection of Gla-3 and c-met is significant to the differentiation of thyroid neoplasm and the evaluation of prognosis.
