【摘要】 目的:研究小鼠胸膜间皮细胞(pleural mesothelial cell,PMC)免疫相关基因表达谱的状况。从而探索用现代免疫及基因芯片技术评价解释中医藏象理论的新方法。方法:取8只BALB/c小鼠胸膜组织块剪切成碎块,提取mRNA,经标记、杂交、清洗、芯片扫描、图象采集与数据分析等过程获得基因表达谱。结果:共筛选出小鼠胸膜间皮细胞免疫相关表达基因204条。其中主要有免疫应答及调节基因56条;炎性反应基因32条;非特异性免疫基因14条;T细胞增殖、活化、调节等相关基因23条,B细胞活化、分化、调节等相关基因9条;急性反应期基因5条;抗原加工与提呈基因4条;Ⅱ型超敏反应正向调节基因5条。结论:胸膜间皮细胞既表达免疫相关的正向调节基因亦表达负向免疫相关的调节基因。提示胸膜可能为一免疫协调器官,这对丰富中医藏象理论与肺气的实质将提供分子生物学的基础研究依据与新的思路。更多还原

【Abstract】 Objective:To investigate the condition of mice pleural mesothelial cell(PMC)immunity-related gene expression spectra, accordingly to explore with modern immunity and gene chip technique to evaluate traditional Chinese medicine ZangXiang theory with the latest method. Methods:DNA chip was used to detect the mRNA from 8 mice tissues of pleural mesothelial cell(PMC). Results:204 immunity- related genes expression were picked out in mice PMC,including 56 immune responses and immune regulatory genes,32 inflammatory reaction genes, 14 non-specificity immunogenes,23 related genes with T cell proliferation,activation,regulation etc; 9 related genes with B cell differentiation,activation,regulation etc; 5 acute reaction period gene,4 antigen processing and presentation genes,5 up-regulation genes of type Ⅱ hypersensitivity reaction.Conclusion:PMC not only expreses immune-associated up-regulation gene,but also expreses immunity-related down-regulation gene. To hint pleura may be one immune coordinator,there will be molecule biological underlaying research identification to be provided for an abundance ZuangXiang theory and lung-QI parenchyma of traditional Chinese medicine,as well as there will be new research thinking to be provided.更多还原
