北方地区黑杨派栽培品种的AFLP指纹分析

【摘要】 以我国北方地区杨树人工林广泛种植的10个黑杨派品种(36杨、50杨、107杨、108杨、111杨、加杨、沙兰杨、I-214杨、109杨和110杨)为实验材料,先进行了苗期形态特征分析,并利用AFLP标记技术,选用E-TA/M-CAG和E-TC/M-CAG两对选择性扩增引物组合,获得了12条特异性条带,建立了这10个杨树品种的DNA指纹图谱,每个品种的扩增带型互不相同,可为杨树品种或无性系鉴定提供独特的特征。结果表明:利用AFLP DNA指纹图谱可以灵敏地鉴别相近无性系并与形态学鉴别分类结论相吻合,表明DNA指纹鉴定配合形态学鉴定方法具有较高的准确性和有效性。

【Abstract】 Ten cultivars of the section Aigeiros widely planted in North China for poplar culture and intensive forest plantations were selected for AFLP fingerprinting,including P.deltoides Bartr.cl ’2KEN8’,P.deltoides Marsh.cl.’Drava’(55/65),P.×euramericana Moench cl.’Neva’(74/76),P.×euramericana Moench cl ’Guariento’,P.×euramericana Moench cl ’Bellotto’,P.×euramericana Moench cl.’Jiayang’,P.×euramericana Moench cl ’Sacrau-79’,P.×euramericana Moench cl ’I-214’,P.deltoides×P.alba cl.’Mincio’,P.deltoides Marsh.× P.maximowiczii Henry cl.’Eridano’.One-year-old plantlets of these ten cultivars were used for morphological analysis in nursery.As the results of AFLP fingerprinting for these ten cultivars,12 polymorphic bands were amplified with two primer combinations E-TA/M-CAG and E-TC/M-CAG respectively.Based on the 12 reliable AFLP polymorphic bands,the DNA fingerprint was constructed to identify these 10 cultivars.In this DNA fingerprint,each genotype had its unique finger pattern and could be easily distinguished from the others.DNA fingerprinting analysis provides a good method for cultivars identification and can overcome the limitations associated with morphological and origin data.
