黄芪多糖对人单核细胞源性的树突状细胞成熟和免疫功能的影响

【摘要】 目的探讨黄芪多糖对人单核细胞源树突状细胞(DCs)成熟和免疫功能的影响。方法将人外周血分离的单核细胞加入含重组人粒细胞巨噬细胞集落刺激因子(20 mg/L)和重组人白细胞介素4(20 mg/L)的培养液中,使其分化为DCs,培养第5天,随机分为空白组(PBS培养液),实验组(黄芪多糖,150 mg/L),对照组(脂多糖,500μg/L),混合培养48 h后,动态观察DCs成熟过程中的形态学变化,流式细胞术检测DCs成熟表型(CD1a、CD80、CD86、人白细胞抗原-DR)。结果与空白组比较,实验组和对照组CD1a、人白细胞抗原DR的表达明显增加(P<0.05),T细胞增殖作用明显增强(P<0.05)。结论黄芪多糖可促进DCs成熟,并增加DCs的免疫活性。

【Abstract】 Objective To study the effect of astragalus polysaccharide(ASP) on the maturation of dendritic cells(DCs).Methods Immature DCs from peripheral blood mononuclear cells were cocultured with ASP(150 mg/L) over night.The cells cocultured with PBS and LPS(500μg/L) served as the negative and positive controls.The morphological changes of DCs were observed serially and the effect of ASP on the maturation of DCs was analyzed by flow cytometer(analying DCs mature phenotypes CD1α,CD80,CD86,HLA-DR).Allogeneic mixed T lymphocytes reaction and supernatant’s cytokine measurement were used for immune functional assay.Results Compared with PBS group,ASP and LPS treated DCs upregulated CD86 and CD1a expression and promoted allogeneic T cells proliferation.Conclusion ASP may play the important role in DC maturation and immune function.
