【摘要】 目的:研究胱冬蛋白酶(caspase)-3、存活素(survivin)在多发性肌炎(PM)/皮肌炎(DM)发病机制中的作用。方法:采用免疫组化技术分析PM/DM患者肌肉组织中caspase-3、survivin表达情况及两者的关系,及其与肌酶学的关系。结果:在47例PM/DM患者中43例caspase-3有不同程度的表达,4例为阴性表达。而survivin全部为不同程度的阳性表达,且与caspase-3有明显的相关关系。在健康对照者的横纹肌中caspase-3表达仅1例,而未见有survivin表达。本研究并未发现caspase-3、survivin表达有性别差异,在PM和DM两组间亦无统计学差异。caspase-3表达与年龄、发病年龄呈负相关,而与肌肉变性程度呈正相关。同时survivin与年龄、发病年龄呈负相关,而与肌肉变性程度呈正相关。结论:caspase-3和survivin是凋亡过程中重要的蛋白,caspase-3和survivin高表达可能在PM/DM发病中起一定作用,而两者又是肌细胞凋亡的重要线索,提示凋亡是PM/DM可能的发病机制之一,但尚需更多的相关研究进一步证实。更多还原

【Abstract】 Objective:To investigate the roles of caspase-3 and survivin in the pathogenesis of polymyositis / dermatomyositis (PM/DM).Methods:Immunohistochemical method was applied to detect the expression of caspase-3 and survivin in the muscular specimens of the patients.Results:In 47 cases of PM/DM,caspase-3 was expressed in different levels in 43 cases,and survivin was expressed in different levels in all cases which was correlated to the expression of caspase-3.Whereas,caspase-3 was detected in only one of the controls,and survivin was not detected in the controls.The expression of caspase-3 was positively correlated to the expression of survivin.The expression levels of caspase-3 and survivin were positively correlated to the extent of muscular degeneration and negatively correlated to the age and the age of onset of the disease of the patients.Conclusion: The expression of caspase-3 and survivin is up-regulated in the muscles of the patients with PM/DM,which may play a role in the pathogenesis of PM/DM.更多还原
