菟丝子醇提液对衰老模型大鼠脾淋巴细胞DNA损伤影响的研究
Effect of alcoholic extract of semen cuscutae on DNA damage in spleen lymphocytes of aging model rats
【摘要】 目的:探讨菟丝子醇提液对衰老模型大鼠脾淋巴细胞DNA损伤的保护作用。方法:Wistar大鼠40只,按体重随机分为青年对照组(8只)、衰老模型组(8只)、菟丝子给药组(24只),菟丝子给药组又分为给药15d、30d和45d组。采用D-半乳糖法制作衰老模型,按0.8g/kg.d剂量经胃灌服菟丝子醇提液。采用单细胞凝胶电泳(SCGE)技术检测脾淋巴细胞DNA损伤程度,同时观察不同给药天数对其影响。结果:衰老模型大鼠脾淋巴细胞DNA拖尾率较青年组明显升高(P<0.01),不同菟丝子醇提液给药组均可降低大鼠脾淋巴细胞DNA拖尾率(P<0.01),以给药45d效果最显著(P<0.01)。结论:菟丝子醇提液对D-半乳糖衰老模型大鼠脾淋巴细胞DNA损伤具有保护作用,且呈现时间依赖性。
【Abstract】 Objective: To study the protective action of alcoholic extract of semen cuscutae(AESC) upon DNA damage in in spleen lymphocytes of aging model rats.Method:Forty Wistar rats were randomly divided into young(8),aging(8),and three different administration groups of 15d,30 d and 45 d(24).Aging rat models were made by D-gal injection,the AESC was drenched by a transgastric approach at the dosage of 0.8g/kg·d.We ssayed the DNA damage in spleen lymphocytes of aging model rat by the single cell gel electrophoresis(SCGE) technique;at the same time,the effects of different days with AESC were observed.Results:Compared with the control group,the percentage of cells with comets of spleen in aging model rat increased(P<0.05);drenched by the AESC the percentage of cells with comets of spleen in aging model rat decreased(P<0.05).Conclusions:AESC can protect DNA damage of the spleen lymphocytes in aging rat induced by D-gal.And the protect presents time dependence.
