不同浓度的骨髓基质细胞裸鼠体内成骨能力的研究
Study on the Effects of Bone Marrow Stromal Cells with Different Seeding Densities in Nude Mice
【摘要】 目的:探讨不同浓度骨髓基质细胞(BMSCs)与胶原膜BME-10X复合培养体的体内成骨能力,为深入开展牙周组织工程研究奠定基础。方法:将BMSCs分别以5×105/mL、5×106/mL和5×107/mL浓度与胶原膜复合培养后植入裸鼠皮下,8周后取材做病理切片,苏木精-伊红、Mallory染色及Ⅰ型胶原免疫组化染色,观察其体内成骨能力。结果:实验组和对照组均有不同程度Ⅰ型胶原的表达,5×106/mL和5×107/mL组高于对照组和5×105/mL组(P<0.05),而5×106/mL与5×107/mL组之间、对照组与5×105/mL组之间无显著型差异。结论:在一定条件下BMSCs的浓度可影响复合体的体内成骨能力,浓度越高其成骨能力越强。
【Abstract】 Objective:To evaluate the osteogenic potential of tissue-engineered composites constructed by collagen membranes(BME-10X) and different concentrations of bone marrow stromal cells(BMSCs) in vivo.Methods:The composites constructed by different concentrations of BMSCs and BME-10X were embedded subcutaneously in nude mice for 8 weeks and then observed by Haematoxylin and eosin(HE) and immunohistochemical staining in typeⅠcollagen,then the average optical intensities were determined and statistically analyzed.Results:Expression of type Ⅰ collagen were detected both in the experimental and control groups.5×106/mL group and 5×107/mL group were significantly higher than blank group and 5×105/mL group(P<0.05),while no signficant differences were found between 5×106/mL group and 5×107/mL group,as well as blank group and 5×105/mL group.Conclusion:The osteogenic potential of cell-collagen membrane composites has cell density-dependence under certain conditions,and the higer concentrations can have stronger activation of osteogenic potentiality in vivo.
