As2O3对人ACC细胞系作用机制的初步研究

【摘要】 目的:探讨三氧化二砷(As2O3)对人唾液腺腺样囊性癌细胞系ACC-M、ACC-2的体外生物学作用及其作用机制。方法:采用形态学观察、MTT法检测As2O3对人唾液腺腺样囊性癌细胞系ACC-M、ACC-2增殖的影响,应用原位末端标记法(TUNEL)及流式细胞仪检测As2O3对ACC-M、ACC-2细胞凋亡作用。结果:As2O3可显著地抑制ACC-M、ACC-2细胞的生长,并且具有时间和剂量依赖性,通过倒置显微镜可明显观察到细胞的形态学改变;通过TUNEL法、流式细胞仪检测显示As2O3可以诱导ACC-M、ACC-2细胞凋亡;并在G2-M期阻滞。结论:As2O3能显著抑制人唾液腺腺样囊性癌细胞的体外生长,其主要作用是诱导细胞凋亡。

【Abstract】 Objective: To investigate the biological effects of arsenic trioxide(As2O3) on human adenoid cystic carcinoma cell line in vitro. Methods: The effects of As2O3 on ACC-M and ACC-2 cell lines were investigate in vitro with morphological method,MTT, TUNEL assay and flow cytometry (FCM). Results: The proliferation of cancer cells were inhibited by As2O3 with a time/dose-dependent manner. As2O3 could induce the cell apoptosis of ACC-M and ACC-2 which revealed by TUNEL and FCM. Conclusion: As2O3 can inhibit the growth of human adenoid cystic carcinoma cell line and induce its apoptosis in vitro.
