帕金森氏病发病率的性别差异及机制的动物实验研究
Gender Difference in Incidence of Parkinson’s Disease and the Possible Mechanism in PD Model
【摘要】 目的通过研究大鼠黑质受损时黑质感觉门控活动、多巴胺神经元数量和大鼠行为学异常变化的性别差异,探讨大鼠的PD发病率是否有性别差异以及可能的发病原因.方法在大鼠右侧黑质埋置记录电极,其中制备PD模型的大鼠同时在右侧黑质微量注射6-OHDA.分别记录各组黑质感觉门控P50;用阿扑吗啡诱发大鼠旋转,检测大鼠的PD发病率,并通过免疫组化测定黑质的TH+细胞数目.结果雌、雄PD模型组损伤侧黑质P50的T/C比值均明显低于正常鼠,且两组大鼠损伤侧黑质TH+细胞数目比各自健侧黑质分别减少;PD发病率雄鼠明显高于雌鼠,切除双侧卵巢的PD大鼠PD发病率明显高于保留卵巢的PD大鼠.结论(1)黑质DA神经元的生存及感觉门控P50活动的水平均和大鼠体内的雌激素水平有关,并且PD的发病率与性别相关,雄性PD发病率明显高于雌性.(2)体内低水平的雌激素在PD的发生中起着重要的作用.
【Abstract】 Objective To study the difference in sensory gating(SG)of SN,the quantity of dopamine neurons of SN,and the abnormal behavior between female and male rats when their SN are damaged,to make sure whether incidence of PD is different between male and female rats and may provide some useful evidence for that.Methods All rats were planted recording electrode in right SN,and the PD rats were injected 6-OHDA in right SN at the same time.Then all rats were recorded sensory gating P50(auditory evoked potential P50,P50),induced rotation behaviour with apomorphine and determined quantity of TH positive cells of SN by immunohitochemistry method.Results T/C ratio in M-PD rats and F-PD rats in damaged side SN was significantly decreased,and the quantity of TH+ cells in control side SN of both group rats was significantly decreased respectively;incidence of PD of F-PD rats was significantly higher than that of M-PD rats,incidence of PD of OVX-PD was significantly higher than that of F-PD.Conclusion(1)The levels of estrogen are related to both the existence of dopaminergic neurons in the SN and level of P50,and there exists sexual differences in the incidence of PD,which in male is higher than female.(3)All above suggest that low levels of estrogen in body is very important in avoiding PD.
