弓形虫RH株和B36株致密颗粒蛋白1基因的克隆和序列分析
Cloning and Sequence Analysis of GRA1 Gene from RH and B36 Strains of Toxoplasma Gondii
【摘要】 目的分析弓形虫2个不同分离株RH株和B36株致密颗粒蛋白1(GRA1)基因的异同.方法应用PCR技术,从弓形虫RH株、B36株分别扩增出致密颗粒蛋白1(GRA1)目的基因片段.将此目的基因片段插入PCR产物克隆专用载体pMD-18-T中,转化大肠埃希菌JM109,PCR鉴定其准确性并测序.DNAstar软件序列分析结果.结果获得弓形虫两个不同分离株RH株、B36株GRA1目的基因片段pMD-18-T/GRA1重组质粒.序列测定表明GRA1目的基因片断与GenBank报道的RH株GRA1基因(注册序列号:EU983103.1)有99%的同源性.经DNAstar5.0软件分析,发现两种虫株GRA1基因序列(R-RH株、B-B36株)有100%的相似性.结论GRA1基因在2个不同分离虫株之间无明显差异,具有较高的保守性.
【Abstract】 Objective To analyze the difference of GRA1 gene from RH and B36 strains of Toxoplasma gondii.Methods The specific fragment of GRA1 gene was obtained by PCR from RH and B36 strains of Toxoplasma gondii.The fragments of PCR products were cloned into a high level sequencing vector pMD-18- T, then transferred into JM109,a recombinant pDM-18- T/GRA1 was constructed,it was identified by PCR.The DNA sequence analysis was done by DNA star software.Results The recombinant pDM- 18- T/GRA1 from RH and B36 strains of Toxoplasma gondii was constructed. After homology analysis,the specific fragment of GRA1 gene from RH and B36 strains of Toxoplasma gondii showed 99% homology with RH strain from GenBank (No. of sequence:EU983103.1).The two specific fragments of GRA1 genes from RH and B36 strains of Toxoplasma gondii shared 100% homology by DNA star software.Conclusions There is no difference in the sequences of the GRA1 gene between from RH and B36 strains of Toxoplasma gondii. The GRA1 gene of Toxoplasma gondii is highly conservative.
