微波灭活兔股骨头坏死模型建立的实验研究

【摘要】 目的探讨用微波灭活制备股骨头坏死动物模型的可行性及技术方法.方法成年新西兰大白兔84只,用微波天线插入兔股骨头内对股骨头灭活10min,根据微波灭活的不同温度随机分为3组.各组分别于术后6个时间点采用X线摄片、MRI扫描和组织学观察股骨头组织的坏死和修复情况,确定最佳灭活温度.结果术后2周所有兔股骨头的MRI扫描都有信号改变;55℃灭活组于4周、8周时X线摄片可见低密度改变和囊性变,组织学检查表明此时坏死与修复同时进行,于术后12周X线摄片有69%兔股骨头塌陷变形.结论采用微波灭活建立股骨头坏死模型是可行的,该造模方法具有动物死亡率低,坏死成功率高,灭活温度和时间容易精确控制,可重复性大,操作简便等优点.55℃为微波灭活制备兔股骨头坏死模型的适宜温度.

【Abstract】 Objective To study the possibility of femoral head necrosis model of rabbits induced by microwave heating.Methods 84 rabbits were randomly divided into three groups according to the microwave heating temperature.Then the microwave antenna was inserted into the center of femoral head to heat for 10 minutes.At six different times after operation,a series of examinations including X-ray,histology and MRI were carried out to observe the femoral head necrosis and repair,and find the optimum microwave heating temperature.Results All the femoral heads of rabbits showed some changes in MRI after 2 weeks;The group of 55 ℃ showed a low density change and cystic degeneration from x-ray photographs,the osteonecrosis and repair occurred at the same time in histology after 4 and 8 weeks.There was 69% femoral heads of rabbits collapsed after 12 weeks.Conclusions It’s feasible to make a new femoral head necrosis (FHN) model of rabbits induced by microwave heating.This method has some advantages including the low animal death rate,the high success rate,controllable temperature and time,good repeatability and manipulation,and so on.55 ℃ is the optimum temperature for the development of femoral head necrosis model of rabbits induced by microwave heating.
