开郁清胃颗粒改善OLETF大鼠胰岛素抵抗机理的研究
Mechanism of Kaiyuqingwei Granules for reliving insulin resistance in OLETF rats
【摘要】 目的探讨开郁清胃颗粒改善OLETF大鼠模型胰岛素抵抗作用的机制。方法32周龄雄性OLETF实验大鼠随机分成4组:模型组、罗格列酮组、开郁清胃颗粒大、小剂量组。正常对照组选用同周龄Wistar大鼠。各组分别给药4周行正常血糖高胰岛素钳夹实验后检测空腹血糖、胰岛素、胰高血糖素、皮质醇、胃动素、胰岛素样生长因子-1(IGF-1)、甘油三酯和胆固醇含量的变化,胰腺切片HE染色观察组织学改变。结果开郁清胃颗粒对OLETF大鼠空腹血糖、胰岛素、胰高血糖素和胃动素无明显影响,但能降低OLETF大鼠的皮质醇、甘油三酯和胆固醇水平;升高胰岛素样生长因子-1水平。OLETF模型组胰岛表现为脂肪变性、炎细胞浸润和纤维增生,开郁清胃颗粒大、小剂量组胰岛结构明显好于模型组。结论开郁清胃颗粒能降低OLETF大鼠的胰岛素抵抗的作用机理与升高IGF-1,降低皮质醇、甘油三酯和胆固醇水平有关,并能有效地保护OLETF鼠胰腺组织形态结构。
【Abstract】 Objective To discuss the mechanism of Kaiyuqingwei Granules(KG) for reliving insulin resistance in Ostuya long-Evans Tokushima fatty(OLETF) rats.Methods Female OLETF rats(32 weeks old) were randomly divided into 4 groups,including the model group,rosiglitazone group,KG group 1(with high dose of KG) and KG group 2(with low dose of KG).The normal control group was set up with Wistar rats(32 weeks old).After 4-week treatment respectively the levels of fasting blood glucose,insulin,glucagon,cortisol,motilin,insulin-like growth factor-1(IGF-1),and contents of serum triglyceride(TG) and cholesterol(CH) were detected in all groups after hyperglycemic clamp test.The histological changes were observed by HE staining of pancreas tissue section.Results KG had no obvious influences on the levels of fasting blood glucose,insulin,glucagons and motilin,but decreased the contents of serum cortisol,TG and CH,and increased IGF-1 level in OLETF rats.In the model group pancreas tissue showed fatty degeneration,inflammatory cell infiltration and fibrosis.In KG group 1 and KG group 2 the structures of pancreas tissue were much better than those in the model group.Conclusion The mechanism of KG for relieving insulin resistance in OLETF rats is related to that it can increase IGF-1 level,and decreased the contents of serum cortisol,TG and CH.KG can protect effectively the histomorphological structure of pancreas tissue in OLETF rats.
