【摘要】 目的研究Survivin在多发性骨髓瘤(MM)中的表达及其与血管内皮生长因子(vascular endothelial growth factor,VEGF)之间的相关性。方法采用RT-PCR和Western blot法检测骨髓瘤细胞系RPMI8226及28例MM患者(其中初治16例、复发/难治12例),6例正常人骨髓细胞中Survivin和VEGF mRNA、蛋白质表达水平。结果骨髓瘤细胞系RPMI8226高表达Survivin和VEGF。Survivin mRNA在正常人、初治和难治/复发MM患者骨髓细胞中阳性率分别为19.8%、77.3%和80.2%,表达水平(Survivin与内参比)分别为0.07±0.03、0.37±0.12和1.2±0.31,MM患者表达率和表达水平显著高于正常人(P<0.01),而难治/复发MM组表达水平高于初治组(P<0.05)。在初治和难治/复发MM组Survivin蛋白阳性率分别为43.8%和61%(P>0.05),难治/复发MM组表达水平明显高于初治组。VEGF在MM中的表达变化与Survivin呈相同趋势。结论Survivin和VEGF参与了MM的发病且与疾病进展有关,两者间存在协同关系。更多还原

【Abstract】 Objective To investigate expression of Survivin and VEGF in myeloma cells and their relationship, and to explore potential interest of survivin as a target in Multiple Myeloma treatment.Methods Reverse transcription-polymerase chain reaction(RT-PCR)and Western blot were used to detect mRNA and protein expression of Survivin and VEGF in multiple myeloma cell line RPMI8226 and bone marrow samples of 16 newly diagnosed MM patients, 12 relapsed /refractory MM patients, and 6 healthy people.Results RPMI8226 cells expressed Survivin and VEGF strongly, The positive rates and the expression levels of Survivin mRNA were significantly lower in healthy people than that of in newly diagnosed patients and relapsed/refractory patients(19.8%vs.77.3%and 80.2%;0.07±0.03 vs.0.37±0.12 and 1.2±0.31,P<0.01),and were significantly lower in newly diagnosed patients than that of in relapsed/ refractory patients(P<0.05).Survivin protein were not detected in all of the healthy people;while the positive rates of Survivin protein were 43.8% in newly diagnosed patients and 61% in relapsed/refractory patientsrespectively(P>0.05),and its expression levels was significantly higher in relapsed/refractory patients than that of in newly diagnosed patients.Also, there were positive relations between VEGF and survivin.Conclusions Survivin and VEGF are all involved in the pathogenesis of Multiple Myeloma, and related with the progression of MM, and they were corporated with each other in MM.更多还原
