“慢性肾衰基本方”抗肾小球硬化的实验研究
The Effect of "Chronic Kidney Failure Basic Prescription" on Glomerulosclerosis in Remnant Renal Rats
【摘要】 目的:探讨"慢性肾衰基本方"对5/6肾切除慢性肾衰竭大鼠模型肾小球硬化的拮抗作用。方法:应用"慢性肾衰基本方"处理构建成功的5/6肾切除大鼠模型4周;应用全自动生化分析仪及血细胞分析仪检测实验动物血清钙(Ca2+)、磷(P3-)、尿素氮(BUN)、血清肌酐(Scr)水平以及血红蛋白(Hb)浓度,HE染色及PASM染色结合图像分析评估系膜/肾小球面积比值,免疫组化染色结合数码医学图像分析系统定量评判肾小球α-平滑肌肌动蛋白(α-SMA)的表达量。结果:"慢性肾衰基本方"治疗组血清BUN、Scr、P3-水平和系膜/肾小球面积比值显著低于模型对照组,而血清Ca2+水平和Hb浓度则显著高于对照组,肾衰竭大鼠系膜/肾小球面积比值和肾小球α-SMA的表达量呈正相关。结论:"慢性肾衰基本方"可减轻肾小球硬化,保护残余肾功能,其机制可能与抑制肾小球固有细胞转分化有关。
【Abstract】 Objective:To investigate the effects of "chronic kidney failure basic prescription" (慢性肾衰基本方)on glomerulosclerosis in 5/6 nephrectomized rats.Methods:5/6 nephrectomized chronic renal failure rats were treated with "chronic kidney failure basic prescription" herbs for 4 weeks.After 4w of treatment,the levels of Hb as well as sera BUN,Scr,Ca2+and P3-were evaluated.The rate of mesangial area/glomerular area(M/G) and the expression of α smooth muscle actin (α-SMA) were analyzed by quantitative analyzed respectively using motic digital medical image-analysis system.Results:Compared with control group,the levels of BUN,Scr,P3-as well as the M/G and the expression of α-SMA in untreated group were significantly increased,while Hb and sera Ca2+ were significantly decreased.Compared with untreated group,the levels of BUN,Scr and P3-as well as the M/G and the expression of α-SMA in therapy group were significantly decreased,while the levels of Hb and sera Ca2+ were significantly increased.Further more,positive correlation between M/G and the expression of α-SMA in glomeruli was found.Conclusion:"chronic kidney failure basic prescription" can alleviate glomerulosclerosis and its possible mechanism is to prevent phenotypic transdifferenciation of glomerular intrinsic cells.
