基于多元曲线分辨-交替最小二乘-紫外光谱监控地黄炮制过程

【摘要】 称取若干地黄样品,用去离子水定容、超声震荡、过滤后测定紫外(UV)光谱。采用多元曲线分辨-交替最小二乘法(MCR-ALS)分析测得的UV光谱数据,提取出3种纯组分的UV光谱,并计算混合体系中其相对浓度。结果表明,从混合组分中提取的UV光谱可以用来将炮制过程中混合组分简单地定性,根据这些组分相对浓度升降的变化趋势,监控地黄炮制过程,确定炮制终点。MCR-ALS-UV法为监控地黄炮制过程及确定炮制终点提供了新途径。

【Abstract】 The UV spectra of several grams of sample of the grinded radix rehmanniae were measured after they were fixed at constant volume by deionized water,ultrasonic vibrated,and filtrated.The spectral data were then analyzed and measured by multivariate curve resolution-alternating least squares(MCR-ALS).Three UV spectral profiles of the components were directly extracted from the mixed data,and their relative concentrations were calculated.The results show that the calculated UV spectral profiles can be used to make brief qualitative confirmation of the components in radix rehmanniae,and the end-point of the processing procedure can be determined by the increasing or decreasing tendencies of the components.MCR-ALS-UV method provides a new approack for monitoring and endpoint determination of the procedure of processing radix rehmanniae.
