颗粒细胞去除方法对猪成熟卵母细胞冷冻效果的影响
Effect of different degranulation methods on cryopreservation of porcine mature oocyte
【摘要】 采集新鲜猪卵巢表面直径2~5 mm的卵泡中卵丘-卵母细胞复合体,采用透明质酸酶消化法和机械法去除成熟卵母细胞外围的颗粒细胞后进行冷冻,解冻后观察其形态正常率和二乙酸荧光素染色存活率,探讨去除颗粒细胞的方法对猪成熟卵母细胞冷冻效果的影响。结果表明:卵母细胞冷冻-解冻后,其形态正常率和存活率均显著降低;酶消化法和机械法去除部分颗粒细胞后的卵母细胞,经冷冻-解冻后形态正常率分别为90.6%和90.2%,两者差异不显著;而存活率分别为49.3%和43.9%,两者显著差异。
【Abstract】 The purpose of this experiment was to explore the effect of different degranulation methods on porcine mature oocytes.Cumulus-oocyte complexes(COCs) were collected from antral follicles(2~5 mm indiameter),and mature oocytes were derived from COCs matured in vitro.We observed the normal rate morphologically and survival rate by FDA staining of frozen-thawed oocytes by different denudation methods.The results showed that the normal rate morphologically and survival rate of oocytes were obviously lower after thawing(P<0.05).The normal rate morphologically were 90.6% and 90.2%,there were no obvious difference(P>0.05),and survival rate were 49.3% and 44.4%(P<0.05),respectively,by enzyme method and mechanical method to denude granulose cells.
