人胚胎脊髓发育过程中脑源性神经营养因子的表达及变化
Changes of brain-derived neurotrophic factor expressions in the human embryo spinal cord
【摘要】 目的:探讨脑源性神经营养因子(BDNF)在不同发育时期人胚胎脊髓中的表达变化及意义。方法:采用免疫组织化学和免疫印迹法对3周~8月人胚脊髓中BDNF蛋白进行定位、定量研究。结果:所检测的15个时段中,神经管上皮和室管膜上皮均可见BDNF阳性细胞,且随胚龄增加呈规律性变化;免疫印迹法显示6~9周,BDNF蛋白含量随胚龄逐渐增加,9周时达到高峰,3月后下降,6月后又有所增加但相对平稳。结论:BDNF在人胚胎脊髓发育特别是胚期脊髓的发育中发挥重要作用,并可能参与神经上皮神经干细胞的分裂增殖。
【Abstract】 Objective:To investigate the expressional changes of brain-derived neurotrophic factor(BDNF)in the human embryonic spinal cord during different development periods.Methods:Immunohistochemistry and Western blot were used to study the localization and quantitation of BDNF protein in the human embryonic spinal cord from the 3rd week to the 8th month.Results:In 15 detected time points,BDNF positive protein was detected in the neural epithelium and ependymal epithelium,and changed regularly as increasing of embryonic age.The results of Western blot demonstrated that the content of BDNF protein was increasing gradually along with the embryonal age from the 6th to the 9th week,reached the peak in the 9th week(P<0.05),began to decrease at the 3rd month(P<0.05),increased again at 6th month and maintained this level.Conclusion:BDNF may play an important role in the human embryonic spinal cord development especially in the early age and may be involved in the division and growth of the stem cells in neurepithelium.
