莲心碱对大鼠脑缺血再灌注损伤脑组织中一氧化氮和一氧化氮合酶的影响

【摘要】 目的:观察莲心碱对大鼠局灶性脑缺血/再灌注损伤后脑组织一氧化氮(NO)含量和一氧化氮合酶(NOS)活性的影响,为莲心碱治疗脑缺血提供实验依据。方法:线栓法阻塞大鼠大脑中动脉2h/再灌注24h制备大鼠脑缺血再灌注损伤动物模型。24只SD雄性大鼠随机分为3组:假手术组、缺血再灌注组和莲心碱(3 mg/kg)组,分别于术前30 min、再灌注即刻、再灌注12h舌静脉注射莲心碱3 mg/kg。再灌注24h后处死大鼠,取脑组织以硝酸还原酶法测定脑组织中的NO水平,以化学比色法测定总NOS活性,并测定诱导型一氧化氮合酶(i NOS)活力。结果:大鼠脑缺血2h、再灌注24h后,脑组织NO水平显著升高,总NOS和i NOS活性显著增加,莲心碱可显著降低缺血再灌注损伤大鼠脑组织NO含量和总NOS、i NOS活力。结论:莲心碱可能通过降低NO对脑缺血再灌注损伤后的神经毒性作用而发挥脑缺血的保护作用。

【Abstract】 Objective:To investigate the effect of Liensinine on levels of NO and NOS in rats with cerebral ischemia-reperfusion injury.Methods:The right middle cerebral artery of the rat was occluded by inserting a thread through internal carotid artery for 2h, and then reperfused for 24h.24 male Sprague-Dawley rats were randomly divided into three groups.sham operation group, ischemia-reperfusion group, Liensinine group.Liensinine (3mg·kg-1) was injected at 30min before ischemia、after reperfusion and 12h,24h after reperfusion, tissue from the forebrain was homogenized, NO and NOS (including total NOS and iNOS) were determined. Results:The levels of NO and NOS levels were significantly increased in the brain tissue of ischemia-reperfusion group compared with shame-operate group.Compared with ischemia-reperfusion group, NO and NOS were significantly decreased in the Liensinine group .Conclusion:Liensinine can reduce ischemia-reperfusion injury by decreasing the damages of NO .
