低聚原花青素对1型糖尿病小鼠血脂代谢和胰岛功能的影响

【摘要】 目的:探讨低聚原花青素(OPC)对1型糖尿病(T1DM)小鼠血脂代谢和胰岛功能的影响及意义。方法:用链脲佐菌素(STZ)小量多次腹腔注射制作T1DM小鼠模型;实验组分3个剂量每天灌胃OPC液,模型对照组和正常对照组则灌胃等容积蒸馏水。4周末酶法检测动物血清总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)和低密度脂蛋白胆固醇(LDL-C)含量;免疫组织化学法检查胰腺组织胰岛中胰岛素蛋白的表达。结果:与模型对照组比较,OPC给药组症状改善;血清TC、TG和LDL-C明显降低(P<0.05,P<0.01);HDL-C有所增加,但不具统计学差异(P>0.05)。胰腺组织免疫组化染色结果显示:模型组胰岛中有多量淋巴细胞浸润,仅少量残留的胰岛细胞被染成棕色,胰岛素表达明显减少;OPC组胰岛中着色的胰岛细胞明显增多,胰岛素表达明显增加,淋巴细胞浸润明显减少。结论:OPC能够降低糖尿病小鼠的血脂含量,提高胰岛分泌功能,减轻由STZ诱导的自身免疫反应,有利于改善糖尿病症状和减少糖尿病心血管并发症的发生。

【Abstract】 Objective:To study the effects of OPC on blood lipid and Islet function in Type 1 diabetes(T1DM) mice.Methods:T1DM was induced in male KM mice with multiple low dose streptozotocin(ld-STZ)by i.p.administration;to treat the T1DM mice with oligomeric proanthocyanidin(OPC)in doses of 50、100and 150mg/kg respectively by oral administration once every day for 4 weeks.Then the content of TC、TG、LDL-C、HDL-C in serum of experiment mice were measured;The expressions of Insulin on islet cells were detected by immunohistochemical Straining.Results:The effect of OPC in doses of 100 and 150mg/kg on reducing serum lipid content in test group was statistically different(P <0.05,P<0.01, espectively )in comparison with DM control group,while the expressions of Insulin on islet cells were obviously stronger than that in test group.Conclusion:OPC has the effects of decreasing the concentration of blood lipid and repairing the injury of islet cells of T1DM mice.
