食管鳞癌p53基因突变和p53功能失活的对照研究
Comparative Study between p53 Gene Mutation and Functional Inactivation of p53 in Esophageal Squamous Cell Carcinoma
【摘要】 目的研究食管鳞癌p53基因突变在p53功能失活中所占的比例,并初步探索食管鳞癌p53功能失活的原因。方法对45例新鲜食管鳞癌组织(正常食管组织作为对照)进行p53基因突变检测(p53-PCR-SSCP法)、p53功能失活检测(p53功能测定法)以及2种p53结合蛋白MDM2和HPV16/18-E6的表达检测(免疫组化SP法)。对上述结果进行统计学分析。结果p53功能失活率为64%,p53基因突变率为49%;p53基因无突变组中,p53功能失活者HPV16/18-E6表达率高于p53功能正常者,也高于p53基因突变组;而3组间MDM2表达率无统计学差异。结论食管鳞癌p53功能失活率高于p53基因突变率,p53功能失活有望成为一种评价食管鳞癌生物学特性的新指标;食管鳞癌p53功能失活的原因可分为p53基因突变和非基因突变原因,前者占72%,后者占28%。在非基因突变原因中可能存在p53结合蛋白HPV16/18-E6的参与,而不存在MDM2的参与。
【Abstract】 Objective To research the proportion of the p53 gene mutation to the functional inactivation of the p53 and seek for the causes of the functional inactivation of the p53 in esophageal squamous cell carcinoma.Methods 45 cases of fresh tissues with esophageal squamous cell carcinoma(the normal tissues were taken as the control) were detected for the p53 gene mutation(p53-PCR-SSCP),the functional inactivation of the p53 protein(the detection of the functional status of the p53 protein) and the expressions of the two p53-associated proteins MDM2 and PV16/18-E6(by immunohistochemical method for SP).Then all the results were statistically analyzed.Results The positive rate of the functional inactivation of the p53 was 64% and the positive rate of the p53 gene mutation was 49%;in the group without the p53 gene mutation,the positive expression rate of HPV16/18-E6 for the functional inactivation of the p53 was higher than that for the normal function of the p53,meantime,higher than that for the group of the p53 gene mutation.There was no significant difference among the 3 groups.Conclusion The rate of the functional inactivation of the p53 was higher than that of the p53 gene mutation and it would be expected to be a new index to evaluate the biological activity of esophageal squamous cell carcinoma;The factors resulting in the functional inactivation of the p53 of esophageal squamous cell carcinoma might include the p53 gene mutation and the non-p53 gene mutation.The former seemed to be 72% and the latter to be 28%.Of the latter,it was possible that p53-associated protein HPV16/18-E6 would join in but the MDM2 wouldn’t.
