术芍易激胶囊对腹泻型肠易激综合征模型大鼠结肠黏膜5-HT和5-HT3R表达的影响

【摘要】 目的观察术芍易激胶囊对腹泻型肠易激综合征(D-IBS)模型大鼠结肠黏膜5-羟色胺(5-HT)和5-羟色胺3受体(5-HT3R)的表达的影响。方法将48只SD雌性大鼠随机分为正常组、模型组、得舒特组和术芍易激胶囊高、中、低剂量组,分别予生理盐水、得舒特、术芍易激胶囊灌胃2周。取大鼠结肠黏膜一块,以免疫组化方法检测5-HT和5-HT3R的表达。结果与模型组比较,术芍易激胶囊各剂量组容量阈值增加(P<0.01),5-HT和5-HT3R的表达明显降低(P<0.01),尤以术芍易激胶囊高剂量组为优。结论术芍易激胶囊能抑制D-IBS模型大鼠结肠黏膜5-HT和5-HT3R的表达,可能通过进一步影响肠道动力学和肠道敏感性来实现其治疗作用。

【Abstract】 Objective To investigate the effects of Zhushao Yiji Capsule(SYC) on the expression of 5-Hydroxytryptamine(5-HT) and 5-Hydroxytryptamine 3 receptor(5-HT3R)in colonic mucosa of diarrhea-predominant Irritable Bowel Syndrome(D-IBS) rats,and to explore its possible underlying mechanisms.Method 48 female rats were randomly divided into six groups:normal group,untreated group,pinaverium treated group,high,medium,low dose of SYC treated group.Each group received 0.9% NaCl,0.9% NaCl,Dicetel(pinaverium),high,medium,low dose of SYC.Immunohistochemical method was used to determine the expressions of 5-HT and 5-HT3R in colonic mucosa of D-IBS rats.Results The expressions of 5-HT and 5-HT3R were significantly lower in the model rats being treated with high dose of SYC than those in the untreated model rats(P<0.05 or P<0.01),but these indexes were insignificantly different between the model rats treated with low,medium dose of SYC and untreated model rats(P>0.05).Conclusion SYC effectively decrease the expressions of 5-HT and 5-HT3R in colonic mucosa of D-IBS model rats.This may be the mechanism of SYC in the treatment of D-IBS.
