姜黄素对巨噬细胞源性荷脂细胞胆固醇水平及caveolin-1表达的影响

【摘要】 目的观察姜黄素对巨噬细胞源性荷脂细胞胆固醇水平的影响,并初步探讨caveolin-1蛋白表达在其中的作用。方法实验分组分为正常巨噬细胞组、巨噬细胞源性荷脂细胞组和姜黄素处理组。3组均采用油红O染色检测细胞荷脂情况,高效液相色谱分析法检测细胞内胆固醇含量,Western印迹法检测caveolin-1蛋白的表达。结果姜黄素作用于巨噬细胞源性荷脂细胞,细胞内总胆固醇、游离胆固醇和胆固醇酯含量均下降;caveolin-1蛋白表达上调。结论姜黄素引起巨噬细胞源性荷脂细胞胆固醇含量下降的同时,caveolin-1蛋白表达上调。

【Abstract】 Objective To observe the effect of curcumin on the cholesterol metabolism in macrophage-derived cholesterol-load cell,and to investigate the effect of the expression of caveolin-1 in this process. Methods Three groups are designed in the experimrnt as follows: the normal macrophage group,the macrophage-derived cholesterol-load cells group and the macrophage-derived cholesterol-load cells group effected by curcumin.Rathonum red stain and high performance liquid phase analytic process are used to assess and detect the level of cholesterol.The expression of caveolin-1 is detected by Western Blot.Results With curcumin effects,the level of total,free and esterified cholesterol decrease in macrophage-derived cholesterol-load cell,while the expression of caveolin-1 is up-regulated. Conclusion Curcumin can degrade the level of total,free and esterified cholesterol in macrophage-derived cholesterol-load cell and it is contributed to the up-regulation of the expression of caveolin-1 at the same time.
