大黄鱼(Pseudosciaena crocea)养殖群体遗传多样性的降低
LOSS OF THE GENETIC DIVERSITY IN CULTIVATED POPULATIONS OF PSEUDOSCIAENA CROCEA BY AFLP
【摘要】 采用AFLP技术方法,对福建二个野生及二个养殖大黄鱼群体进行了遗传多样性的研究。实验采用7对引物组合,在四个群体的120个个体中共扩增出472个位点,其中多态位点为220个,多态位点比例为46.61%。结果表明,大黄鱼养殖群体的遗传多样性降低:宁德野生群体和湄州野生群体多态位点比例分别为42.42%和45.14%,Nei遗传多样性指数分别为0.1046和0.1245,Shannon多样性指数分别为0.1659和0.1942,平均有效等位基因数(Ne)分别为1.4153和1.4428;宁德养殖群体和连江养殖群体多态位点比例分别为40.83%和40.53%,Nei遗传多样性指数分别为0.1027和0.1039,Shannon多样性指数分别为0.1615和0.1633,平均有效等位基因数(Ne)分别为1.3919和1.3856。四个群体间遗传距离为0.0359—0.1465,平均为0.079,群体间基因流为1.0808。
【Abstract】 Genetic diversity of two wild and two cultivated populations of Pseudosciaena crocea in Fujian Province were analyzed by AFLP. A total of 472 loci were detected from 120 individuals in the 4 populations, of which 220 (46.61%) are polymorphic. The results from AFLP technique show that the genetic diversity in the cultivated populations has been reduced compared to that of wild populations. The percentage of polymorphic loci, the Nei’s Gene Diversity, Shannon’s Information Index and the mean effective number of alleles per locus in two wild populations are 42.42%—45.14%, 0.1046—0.1245, 0.1659—0.1942 and 1.4153—1.4428, respectively, which in two cultivated populations are 40.83%—40.53%, 0.1027—0.1039, 0.1615—0.1633 and 1.3919—1.3856, respectively. Genetic distance among the 4 populations varies from 0.0359—0.1465, in the mean of 0.079, and the gene flow (Nm) is 1.0808. These results show a lower-level genetic diversity compared to other fishes.
