三氧化二砷对结肠癌凋亡及耐药基因表达影响的研究
Study of apoptosis and MRP mRNA expression induced by arsenic trioxide on colonic carcinoma cells
【摘要】 目的研究三氧化二砷(As2O3)对结肠癌SW620的抑制和诱导凋亡作用及对MRP基因表达的影响。方法选用结肠癌SW620细胞系,运用流式细胞术检测As2O3对结肠癌的抑制和诱导凋亡作用;用逆转录-聚合酶链反应(RT-PCR)方法检测MRP mRNA的表达。结果As2O3对结肠癌SW620细胞具有抑制生长作用及增加凋亡诱导率,不同浓度的As2O3均可诱导凋亡。1.0μmol/L、2.0μmol/L的As2O3可下调MRP mRNA的表达。结论AsO具有诱导细胞凋亡的作用,并能下调MRP mRNA表达。
【Abstract】 Objective To study the apoptosis and MRP mRNA expression induced by arsenic trioxide(As2O3) on colonic carcinoma cells.Methods The flow cytometry and RT-PCR tests were used to investigate of apoptosis levels induced by arsenic trioxide and the effect on MRP mRNA expression.Results As2O3 inhibited the growth of SW620 colon cancer cells and increased the rate of apoptosis.1.0 μmol/L and 2.0 μmol/L arsenic trioxide decreased MRP mRNA expression.Conclusion Low concentrations may improve apoptosis of colonic cancer as well as decrease MRP mRNA expression.
