喉鳞癌中ΔNp63和ΔNp73的表达及临床意义

【摘要】 目的通过研究喉鳞癌中ΔNp63和ΔNp73的表达及与患者各临床病理参数的关系,探讨其在喉鳞癌发生、发展中的作用。方法采用免疫组化二步法,检测40例喉鳞癌及相应癌旁组织中ΔNp63和ΔNp73的表达情况。结果ΔNp63和ΔNp73在喉鳞癌组织中的阳性表达率分别为85%(34/40)和82.5%(33/40)。喉癌组织中ΔNp63和ΔNp73的表达显著高于相应癌旁组织(P<0.001)。ΔNp63在喉癌组织中的表达与肿瘤分化程度有关(P<0.05);而与患者性别、临床分期、有无淋巴结转移等无关。ΔNp73在喉癌组织中的表达与患者性别、肿瘤分化程度、临床分期、有无淋巴结转移等无关(P>0.05)。喉癌组织中ΔNp63和ΔNp73的表达无显著相关性(r=0.16,P>0.05)。结论ΔNp63和ΔNp73可能作为癌基因维持肿瘤细胞干细胞样的增殖特性,在喉鳞癌的发生、发展中发挥重要作用,并可能成为喉癌成瘤机制研究中的重要靶点。

【Abstract】 Objective To investigate the expression of ΔNp63 and ΔNp73 and their association with clinical pathology characteristics in laryngeal squamous cell carcinoma(LSCC).Methods Expression of ΔNp63 and ΔNp73 was detected by immunehistochemistry in 40 cases of primary LSCC and matched adjacent normal tissues.Results The positive rates of ΔNp63 and ΔNp73 were 85.0% and 82.5%,respectively in laryngeal tumors.The positive rates and expression level in primary LSCC were remarkably higher than that in matched adjacent normal tissues(P<0.001).The expression of ΔNp63 was not correlated with gender,clinical stage and metastasis in lymphoid nodes,however it was correlated with tumor differentiation(P<0.05).The expression of ΔNp73 was not related to gender,tumor differentiation,clinical stage and metastasis in lymphoid nodes(P>0.05).Expression of ΔNp63 and ΔNp73 was not in a correlation in primary LSCC(r=0.16,P>0.05).Conclusion ΔNp63 and ΔNp73 are dominant isoforms and may act as oncogene in LSCC.ΔNp63 and ΔNp73 may be the key targets in research on tumorigenesis of LSCC.
