HPLC同时测定兰石草中的齐墩果酸和熊果酸

【摘要】 目的建立同时测定兰石草中齐墩果酸和熊果酸含量的方法。方法采用HPLC法,色谱柱为Hypersil ODS2(250 mm×4.6 mm,5μm),流动相为甲醇-水(86:14),流速为0.8 ml.min-1,检测波长为215 nm,进样量为10μl,柱温为25℃。结果两种成分均达到基线分离,齐墩果酸和熊果酸的线性范围分别为10~400 ng(r=0.9999),18~720 ng(r=0.9998);平均回收率分别为99.25%(RSD=0.63%,n=5)和97.59%(RSD=1.57%,n=5)。结论所建方法简便、快速,结果准确、可靠。

【Abstract】 OBJECTIVE To establish a quantitative method for the simultaneous determination of ursolic acid and oleanolic acid of Lancea tibetica Hook.f.et Thomas.METHODS HPLC method was adopted.The samples were separated with the column of Hypersil ODS2(250 mm×4.6 mm,5 μm) which was eluted with methanol-water(0.04 % phosphoric acid) at the ratio of 86:14 with detected wavelength at 215 nm.The flow rate was 0.8 ml·min-1 and the column temperature was 25℃.RESULTS The results indicated two compounds were basically isolated.The linear ranges of oleanolic acid and ursolic acid were 10-400 ng(r=0.9999)and 18-720 ng(r=0.9998),respectively.The average recoveries were 99.25 % with RSD of 0.63% and 97.59% with RSD of 1.57%,respectively.CONCLUSION The method is rapid and precise.
