南海海绵Iotrochota sp.中抗氧化活性成分的研究

【摘要】 目的研究南海海绵Iotrochota sp.中的抗氧化活性成分。方法以95%乙醇浸提得南海海绵总浸膏,经多步分配得乙醇、正己烷和正丁醇部位。采用DPPH法进行活性示踪、反相柱层析和HPLC法分离,光谱方法进行结构鉴定。结果分离得到的化合物鉴定为purpurone,其清除DPPH自由基的IC50为19μg.m l-1。结论Purpurone为海绵Iotrochota sp.的特征性成分和主要抗氧化活性成分,对Iotrochota属海绵的分类具有重要意义。

【Abstract】 OBJECTIVE To investigate the antioxidant constituents of marine sponge Iotrochota sp.from South China Sea.METHODS The sponge was extracted with 95% alcohol,and then the crude was partitioned repeatedly between different organic solvents.Guided by the DPPH assay,reversed-phase column chromatography and HPLC were used to separate the chemical constituents in the n-BuOH extract.The structure of the active compound was determined by spectral method.RESULTS One compound was isolated and identified as purpurone.Its IC50 of scavenging DPPH radical was 19 μg·ml-1.CONCLUSION Purpurone is the predominant antioxidant constituent.As a characteristic constituent,it’s very significant for the taxonomy of genus Iotrochota.
