莨菪亭祛暑功效及其机制的实验研究
Experimental Studies on the Effect and Mechanisms of Reducing Summer-heat of Scopoletin
【摘要】 目的研究莨菪亭(Scopoletin)祛暑功效及其机制。方法采用人工湿热环境热应激法复制家兔中暑动物模型,观察莨菪亭对热应激家兔平均发热曲线、到达42℃的时间均数及体温平均上升速度,研究其祛暑功效;应用比色法和滴定法测定家兔热应激1h后,心、脑、肾等组织中Na~+-K~+-ATPase的活性以及肺组织和血清中PLA2的活性,研究其祛暑机制。结果莨菪亭能显著延迟家兔在人工湿热环境中肛温到达42℃的时间(P<0.01~0.001);且明显降低单位时间肛温上升速度(P<0.001);于热应激前给予莨菪亭能有效地抑制心、脑、肾组织中的Na~+-K~+-ATPase活性,以及明显抑制血清、肺组织中的PLA2活性,这些作用与用药剂量呈明显的正相关(P<0.01)。结论莨菪亭通过抑制心、脑、肾组织中的Na~+-K~+-ATPase活性和血清、肺组织中的PLA2活性来发挥祛暑功效
【Abstract】 Objective To study the effect of reducing summer-heat and its mechanisms of Scopo- letin.Methods The temperature changes of heat-stressed or normal rabbits were observed after intragas- tric administrating Scopoletin.At the same time,the activity of Na~+-K~+-ATPase in heart,brain and kidney,the activity of PLA2 in serum and lung were measured by colorimetry or titration.Results Sco- poletin could delay the time of reaching 42℃(P<0.01~0.001) and lower the rising rate of the core tem- perature (Trr) (P<0.001) in heat-stressed rabbits in a dose-dependent manner respectively.It also could inhibit significantly the activity of Na~+-K~+-ATPase and PLA2 in the Heat-stressed rabbits(P<0.01). Conclusion:Inhibited the activity of Na~+-K~+-ATPase in heart,brain,kidney and the activity of PLA2 in lung,serum were the mechanisms of reducing summer-heat effect of Scopoletin.
