电针对心肌梗死后抑郁大鼠模型海马神经元作用机制的研究
Mechanism of electro-acupuncture on hippocampus neurons after myocardial infarction in rats with depression
【摘要】 目的观察电针对心肌梗死后抑郁大鼠模型海马神经元的影响,并探讨可能的作用机制。方法建立心梗后抑郁症大鼠模型,采用电针干预的方法,观察大鼠的体质量和行为学变化。用TUNEL法和免疫组化检测海马神经元细胞凋亡,Western-blotting检测P-CREB。结果心梗后抑郁大鼠体质量、敞箱实验得分、糖水消耗和纯水消耗显著减少,大鼠海马CA3区出现神经元细胞凋亡,P-CREB水平降低。电针干预3周后,与模型组比较,大鼠体质量增加,行为学指标改善,神经元细胞凋亡数减少,Bcl-2表达增加,Bax表达减少,二者比值增高,并P-CREB水平增加,差异均具有统计学意义(P<0.05)。结论心梗后抑郁与海马神经元细胞凋亡和P-CREB水平降低有关,电针刺激可能涉及某些信号通路激活磷酸化CREB,抑制凋亡基因,促进抗凋亡基因的产生。
【Abstract】 Objective To observe the effect of electro-acupuncture(EA) on the hippocam-pus neurons in rats with depression after myocardial infarction(MI),and investigate the possible mechanism on it.Methods The animal model of depression after MI was established and treat-ed with EA.All rats were tested on weight and ethology.The neurons apoptosis of hippocampus were examined with TUNEL and immunohistochemicstry.The level of p-CREB was determined with Western-blotting.Results The weight of rats,score of open-field test,consumption of su-crose solutions and the pure water were decreased significantly.And there were increased neu-rons apoptosis in CA3 and decreased P-CREB in the hippocampus.Compared with model group after three weeks treated with EA,the weight and the ethology of rats were increased corre-spondingly,neurons apoptosis and Bax decreased,and Bcl-2,Bcl-2/Bax and P-CREB increased (P <0.05).Conclusion Depression post-MI may related to decrease the volume of neurons apoptosis and P-CREB in the hippocampus.The possible mechanism of EA stimulation may involved in the change of some signal pathway,inhibition of apoptosis gene and generation of antiapoptosis gene.
