微波法选育红曲霉菌种发酵生产生理活性物质的研究
Study on Microwave Breeding Monascus Ruber and Production of Monacolin K with Fermentation
【摘要】 目的研究红曲霉菌种发酵生产生理活性物质Monacolin K的条件。方法用微波法选育以红曲霉No.1为出发菌,经50W的微波辐照诱变获得Monacolin K高产菌株,并对其产色素和Monacolin K的发酵条件进行摸索和研究。结果接种剂量3 ml、粳米与豆粕的配比=5∶1、培养温度28℃、培养时间9 d,产品色素含量最高达348μg/g、培养11 d Monacolin K达最大值90μg/g。结论通过微波诱变红曲霉菌种,在适当的发酵条件下,可提高次生代谢物Monacolin K的产量,红曲色价变化不大。
【Abstract】 Objective The conditions were studied to product Monacolin K with Monascus ruber′s fermentation.Methods A high-production Monacolin K strain was obtained through 50 W 1.5 min microwave mutation as compared with the original Monascus ruber No1.The fermentation conditions of the strain to product pigment and Monacolin K were searched and studied.Results The results showed that the content of Monacolin K could reach 90 μg/g fermentation in 11 d,and the content pigment was 348 μg/g under fermentation in 9 d,under the optimum fermentation conditions,the 3 ml,and the ratio of the solid culture medium was made of plump rice and soybean meal 5∶1,temperature 28℃.Conclusion By the way of microwave inducing monascus ruber’s fermentation,the yield of Monacolin K can be increased on the appropriate conditions,but the pigment output is immovable.
